And we believe that Fructus Corni polysaccharides is one of the principal active ingredients. Therefore, clearing its chemical structure and content, determining its preparation technology and quality control standard, can further reveal Fructus Corni′s medicinal using value and developing potential. And which provide necessity of basis for Fructus Corni of development.Material and method:In thia article, mature drying Fructus Corni pulp is raw material. We get the polysaccharide by water extraction and alcohol precipitation′s method. To determine the optimum extraction and alcohol precipitation′s process. To establish Fructus Corni polysaccharide content measure method. We use column chromatography to separate the extracted polysaccharide. And study its monosaccharose composition. Clear the types of its monosaccharide composition and proportion. In the last, we conduct the extracted polysaccharide on the in-vitro antioxidant and in-vivo immune experimental test. And clear its pharmacological effect.Conclusion:1. Through orthogonal experiment, I determine the optimum extraction craft. The optimum extraction conditions of the Fructus Corni polysaccharide: the times of extraction was 3, extraction temperature was 100℃, adding 30 folds of water every time, extraction time was 4 hours.2. With the optimum extraction conditions, carry on assaying towards being 22 different places Fructus Corni polysaccharide. The result manifest: the content of different places Fructus Corni polysaccharide is quite different. Among them, Liaoning Yingkou has the highest content, which is 6.22%; Beijing has the lowest contont, which is 1.68%. The polysaccharide content was determinated with phenol-sulfuric acid method. This method is simple, fast and accurate.3. Fructus Corni polysaccharide extracted in water and sink in alcohol, through DEAE-cellulose DE-52 column chromatography gradient separation to get five different parts of A, B, C, D, E. And adopt SephadexG-100 column chromatography gradient depurate A, to get Fructus Corni polysaccharide A′. Through SepharoseCL-6B gel column chromatography to verify the purity of A′. phenol-sulfuric acid method tracks detect, the result indicated that Fructus Corni polysaccharide A′is a uniform polysaccharide. Through lamella analysis, this polysaccharide is composed of glucose, xylose and rhamnose. Chemical composition analysis by TLC and HPLC showed that it was composed of glucose, xylose and rhamnose in a molar ratio of 1:1.77:1.31 respectively.4. Through the in-vitro and in-vivo pharmacological experiment, the result indicate that Fructus Corni polysaccharide has in-vitro antioxygen activity, and can increase small mouse immune activity, depress the blood suger of the diabetes mice. |
