Objective: Using modern technology of membrane filtration to prepare ginseng oral liquid in order that it could solve the problem, such as environment pollution and turbid liquid etc, consisting in the traditional methods of oral liquid preparation. And study on ergonomy of ginseng oral liquid using pharmacological test model to lay a foundation for the development of ginseng oral liquid those quality can be controlled and which comply with the international standard.Methods: To extract ingredient from Radix ginseng by method of aqueous homogenate with different extraction span,extraction times and material/liquid ratio. Using isolation technology of hollow fiber membrane instead of ethanol precipitation step in traditional processing technique of oral liquid to microfiltrate and ultrafiltrate the ginseng extract followed by the HPGFC test of purification.. To optimize the extraction technology of ginseng liquid using orthogonal test with which the extraction yield of ginsenoside and ginseng polysaccharides are the evaluating indicators. Finally using models of mice swim, mice hypoxia and DNFB induced mice delayed hypersensitivity to select the activities of this ginseng oral liquid.Results: The orthogonal test resulted in that among the factors of extraction, extraction span, extraction times and material/liquid ratio all had something to do with the yield of ginsenoside and ginseng polysaccharides. When selecting the one time extraction, the yield would become bigger along with the material/liquid ratio become smaller. When selecting multi-times extraction, extract three times and 24 hours each time with material/liquid ratio of 1:3 would get the greatest yield of ginsenoside and ginseng polysaccharides. By comparing the results of HPGFC after processing the operation of hollow fiber membrane we could draw the conclusion that the non-drug-effect impurity had been effectively removed. The results of pharmacology experiments revealed that ginseng oral liquid could prolong the life span of mice suffering swimming as well as hypoxia and had effect on viscera weight ratio in DNFB induced mice delayed hypersensitivity. All of the effects above had dose-effect relationship.Conclusion: The best extraction process of ginseng oral liquid was of extract one time for 24 hours with material/liquid ratio of 1:7. The ginseng oral liquid being operated under that process had higher yield of ginsenoside and ginseng polysaccharides; the experimental period of it was comparatively short; the purity was good; the active gradients were well-distribute; no leavings; no sediments after being shaked. It suits the situation of modern manufacture for it solved the traditional problems of complex operation, high costs, bad clarity, leaving sediments and solid preparation-induced coarse, big and black impurity etc. Meanwhile it was proved to be effective in enhancing immune system, antifatigue and antihypoxic by prolonging the life span of swimming, oxygen-poor mice and increasing the ear dwelling, thymus/weight ratio and spleen/weight ratio.
