| Purpose: To explorer the molecular mechanism of removing blood stasis by promoting circulation of astragali-scoropendra antifibrosis formula on hepatic injury and provide a theoretical basis for the therapeutic principle of removing blood stasis by promoting circulation.Methods: Hepatic injury model was induced by immunologic injury on 40 rats, then the rats were randomly divided into four groups, 10 Rats serving as control group received normal saline intraperitoneal injection; control group and 10 rats serving as model group received gastric drenching with normal saline; 10 rats serving as high-dose group and 10 rats serving as low-dose group received gastric drenching with traditional Chinese drugs. after 8 weeks continuous medication, we measured weight of spleen, computed weight index of spleen, measured whole blood viscosity, observed the pathological morphous of hepatic tissue, measured hepatic function parameters(ALT,A,G,A/G )by full-automatic biochemical analyzer, measured transforming growth factor-β1 in serum by enzyme linked immunosorbent assay(ELISA), measured the content and expressivity of TGF-β1, Smad2, Smad3, Smad7 by immunohistochemical method(SP method) ,measured the mRNA expressivity of TGF-β1 in hepatic issue by RT-polumerase chain reaction(RT-PCR).Results: In the high-dose group and low-dose group: compared with model group,⑴the weight and weight index of spleen were significantly decreased(p<0.01);⑵whole blood viscosity was significantly decreased(p<0.01);⑶the degeneration, necrosis, fibroplasia of hepatic cells were significantly abated in a dose-dependent manner;⑷hepatic function parameters were significantly improved (p<0.05 or p<0.01);⑸TGF-β1,Smad2,Smad3 in serum and issue were significantly decreased in a dose-dependent manner(p<0.01), Smad2 were significantly increased in a dose-dependent manner(p<0.01);⑹mRNA expressivity of TGF-β1 was decreased( p<0.05).Conclusion: Astragali-scoropendra formula has anti-fibrosis effect on the liver of Rats, the mechanism might be:⑴inhibiting the mRNA expression of TGF-β1 and decreasing TGF-β1 in serum;⑵regulating Smad system;⑶decreasing whole blood viscosity and improving hepatic microcirculation... |
