Scotch pine (Pinus sylvestris L. var. mongolica Litv.) , which is a kind of perennial evergreen arbor,is belongs to pinaceae and pinus. Scotch pine contains abundant volatile oil as other most plants of pinaceae. There is a wide perspective in the study on chemical compositions of the volatile oil.The volatile oil of scotch pine was analyzed by GC/MS and methods of chemometrics qualitatively and quantitatively.To optimize the extracting effective conditions of the volatile oil from scotch pine, the central composite design-response surface methodology has been used. The volatile oil of scotch pine was analyzed qualitatively and quantitatively, and the tendency of chemical compositions were inspected according with months. The chromatogram fingerprint of the oil was established and evaluated. To inspect chemical compositions in the volatile oil of oleoresin, barks, xylem, pine cone and masson pine branchlet-tip, the GC/MS database has been used.The pine needles of scotch pine in the same batch was collected, and the volatile oil was extracted by water distillation method. The soaking time, water addition and extraction time were independent variables and the extraction rate was induced variable. Linear and nolinear mathematic models were used to estimate the relationship between independent and dependent variables. Response surface methodology was used to optimize the process of extraction. The pine needles of scotch pine from Junuary to October were collectred, extrected by the water distillation method at best conditions and detected the oil by GC/MS. Heuristic evolving latent projections (HELP) was used to deal with the data and got the various tendency of the volatile oil in the pine needles of scotch pine from different months. The pine needles of scotch pine from ten areas in Hei Long-jiang province in August were collected. The volatile oil was detected by GC/MS after extraction. Common pattern was established by methods of median and medium value, and the similarity was evaluated. The volatile oil from oleoresin, barks, xylem, pine cone and masson pine branchlet-tip were extracted, and inspected the different chemical compositions.Using the central composite design-response surface methodology and according to the practice, the best extracting conditions were determinated.The sample is extracted with 12.88 times water for 8 h after 12 hours soaking. A mathematic model was established with the multiple correlation (r = 0.935 8 ). In the volatile oil from different months, the number of main compositions varied from 57 to 76. 49 common compositions that relative percentages were more than 0.1% were found, which contented 10 monoterpenes and 27 sesquiterpenes.All of related coefficients between common pattern and TICs of the volatile oil from the different place were more than 0.9. Sample 1 from Da Xing An Ling was different from others. There are 41 main compositions of volatile oil in turpentine, 67 in barks , 50 in pine xylem, 36 in pine cone and 68 in masson pine branchlet-tip.Extraction conditions were optimized by central composite design-response surface methodology, and got a better extraction rate at certain conditions. The multiple correlation of binomial model was so ideal that could response the results better. 20 kinds of compositions were main compositions of the volatile oil, average relative percentage of which were more than 1.0 %. Contents of some main compositions varied with seasons. The method of establishing the chromatographic fingerprint was accurate.Samples and instruments were stable,and the results were effective. Accuracy,precision and stability of chromatographic fingerprint of the volatile oil in scotch pine met the requirements and the chromatographic fingerprint can be used as a QS(Quality Standard). The volatile oil from turpentine, barks, xylem, pine cone and masson pine branchlet-tip were different on chemical composition.Chemical compositions of the volatile oil in pine needles of scotch pine were compared with each other, QS of the volatile oil in pine needles of scotch pine was established, and the basic datas were provided to medical value of the volatile oil. |
