Geniposide, a typical compound of iridoid glucoside, isolated from Fructus Gardeniae, had shown abundant activities such as liver-protective activity, cholagogue activity, anti-inflammatory activity, anti-tumour activity, anti-hypertension activity. In addition, geniposide also showed anti-mutagen activity, antioxidant activity, anti-thrombus activity, neuron-protective activity, repairing the injured tissues and hypoglycemic activity. Hypoglycemic effect in particular, has attracted more attention in recent years. The results show that geniposide mostly achieves its biological activities by genipin which is its aglycone.First, we examined the distribution of geniposide in every part of the fruit of Gardenia jasminoides Ellis by HPLC analysis, which indicated that geniposide mainly contains in the nutlet, and less in the peel.Second, we used Herbal Blitzkrieg Extractor (HBE), a novel extract method, to improve the extraction technology of emulsin from Semen Armeniacae Amarum for the first time, and optimized the new technology. The optimum extraction conditions are 50.0g the powder of defatted Semen Armeniacae Amarum,0.0625mol/L pre-cooling ammonia (500mL+450 mL), extracted twice (120s+90 s), filtrated by gauzes and collected the filtrate. Adjusted pH4.0 by acetic acid, filtered hybrid protein, precipitated the emulsin by 4 times the amount of ethanol, filtration, washed, frozen dried. And we determined the specific activity of emulsin by DNS.Third, we used the emulsin which had extracted to hydrolyze geniposide, and a four-factor five-level quadratic general rotary unitized design was used to optimize the hydrolysis process. Take the hydrolysis rate as the target to examine four major factors, the buffer solution pH, reaction temperature, reaction time and emulsin amount, on the impact of the process. To identify more appropriate conditions are the buffer solution pH5.25, the reaction temperature 50.5℃, the reaction time 90min and emulsin amount 10. And we identified the hydrolysis products of genipin by physical and chemical properties, purity testing and identification of NMR.Finally, we contrastively studied the hypoglycemic activity of geniposide and genipin on alloxan induced animal blood sugar-model. The results showed that, in a certain dose range, both geniposide and genipin were able to improve the survival rate of mice, reduce the blood glucose levels of diabetic mice, and had hypoglycemic effect.
