| Objective:To evaluate the susceptibility of Neisseria gonorrhoeae to five antimicrobial agents in Changzhi of Shanxi.To understand Neisseria gonorrhoeae of SpectinomyCin-resistant and 16S rRNA gene mutations and the relevance of the distribution of the mutant.Methods:(1)Disk diffusion method on thirty-one N.gonorrhoeae was used to test susceptibility of antimicrobial agents including penicillin, tetracycline, ciprofloxacin, ceftriaxone, spectinomycin. (2)The 16S rRNA gene of spectinomycin-resistant Neisseria gonorrhoeae was amplified by PCR.(3)The amplicons were directly analyzed by DNA sequencing.Results:(1)The positivity rates of resistant isolates to penicillin, tetracycline, ciprofloxacin, ceftriaxone, spectinomycin were 57.2%,87.5%,100%,0%,.6.25%. Positivity rates of multiple-resistant isolates to five antimicrobial agents were 96.8%. (2) Two spectinomycin resistant strains and Moderate sensitive strains in the three strains exist in 16S rRNA gene mutation, mutation mainly in the form are in units of 991 point mutation occurred, from A (adenine) was mutated into G (guanine).3 in the Moderate sensitive strains are still 1308 missing A (adenine).Conclusions:(1) Shanxi Changzhi of Neisseria gonorrhoeae to spectinomycin resistance rate and the rate of growth of multi-drug resistance rapidly, ceftriaxone and spectinomycin in treatment of gonorrhea in this region may act as a drug of first choice. (2) Neisseria gonorrhoeae 16S rRNA gene mutations in spectinomycin resistance plays an important role. (3) 16S rRNA gene of Neisseria gonorrhoeae to spectinomycin mutation is an important mechanism for drug resistance. |
