| Sinopodophyllum hexandrum (Royle)Ying which belong to Berberidaceae is the Podophylloideae's perennial herbaceous plant, its root and rhizome contains a lot of podophyllotoxin and its derivatives. In recent years,The podophyllotoxin can be used as precursor substances to synthetized anti-cancer drugs VP-16 (etoposide) and VM-26 (teniposide) for the high antitumor activity.At present, there are mainly contain lignanoids, flavonoids, saponins, polysaccharide and tannic acid in Sinopodophyllum hexandrum (Royle)Ying.The lignanoids composition mainly forpodophyllotoxin,4'-Demethylpodophyllotoxin,Picropodophyllin,dehydropodophyllotoxin,p odophyllotoxone,a-Peltatin,β-Peltatin,Podophyllotoxin-glucoside,4'-Demethylpodophyllotoxi n-glucoside,a-Peltatin-glucoside,β-Peltatin-glucoside,Diphyllin etc.Flavonoids ingredients for quercetin and kaempferol etc.In this project,we mainly selected Sinopodophyllum hexandrum (Royle)Ying as the research object,then the podophyllotoxin have been analysed from the angle of pharmaceutical chemistry,which was extracted from different parts of Sinopodophyllum hexandrum (Royle)Ying in different areas. Based on high performance liquid chromatography(HPLC), the HPLC fingerprint was established.Application of high perfor-mance liquid chromatogrammphy-electrospray ionization trap mass spectrometry(HPLC-ESI-MS) to determine and analyse the quality and the genetic relationship among Sinopodophyllum hexandrum (Royle)Ying varieties.1. Based on the literatures, the suitable chromatographic analysis conditions was selected to analyse and determine the podophyllotoxin which was extracted frome six wild Sinopodophyllum hexandrum (Royle)Ying root and rhizome samples.The results show that the content of podophyllotoxin in root between 4.512-9.834‰and in rhizome between1.218-7.770%o.Besides,The results also show that podophyllotoxin is defferent in different parts of Sinopodophyllum hexandrum (Royle)Ying in different areas,the highest is from Ningxia Liupanshan,then Gansu xinglongshan,Sichuan yulinxiang,Gansu chagang,Sicuan shuajingsi etc.However,the lowest is in Yunnan napahai. In rhizome,the highest is from Yunnan napahai, then is Sichuan yulinxiang,Gansu xinglongshan, Sicuan shuajingsi is the lowest.2.The methanol extracts of Sinopodophyllum hexandrum (Royle)Ying powder were analyzed by RP-HPLC on an ODS column, at 30℃, using a gradient elution with a water(contains 0.04% methanoic acid)-acetonitrile mixture at 0.5 ml-min-1. The eluate was monitored by UV absorption at 290nm.The HPLC fingerprints of Sinopodophyllum hexandrum (Royle)Ying from different areas were established and the correlation coefficients were calculated. Furthermore, the peak of podophyllotoxin was identified. At the same time, the relative content of common peaks were calculated according to the content of podophyllotoxin.3.Application of HPLC and HPLC-ESI-MS connected technology,the suitable chromatographic analysis conditions was selected to analyze and determine the solution which was extracted frome six wild Sinopodophyllum hexandrum (Royle)Ying. LC-ESI-MS(Thermo Scientific,USA) has been used.The chromatographic analysis conditions can be showed as follows:Diamonsil C18(250x4.6mm,5μm) chromatography column,the flow rate was 0.2 ml·min-1,detecting wavelength was at 290 nm and using a gradient elution with a water(contains 0.02% methanoic acid)-acetonitrile mixture, the injection volume was 10μl;ESI ionization source, positiveion mode,ISpray Voltage(kV):4.00,Sheath Gas Flow Rate(arb):30.00,Aux Gas Flow Rate(arb):5.00,Capilitary Temp(℃):300,Tube Lens(V):80,the range of scanning the electronic charge-mass ratio:100-1300m/z.As results,13 components were separated by this method, and then conbine with the relative literatures,them were determined. |
