| Objective To study the chromosome 3,6,8 of choroidal melanoma, determine the pathology of choroidal melanoma,. Analyze the correlation of chromosome and pathology, tumor basal diameter, tumor thickness in choroidal melanoma.Methods From April 2004 to December 2009,36 cases in the ophthmology department of EENT to Fudan University were selected. All the cases were confirmed choroidal melanoma by pathological examination after enucleation. Based on the pathology of modified-chandler classification, the cases were divided into three groups, which include spindle cell type, mixed cell type and epithelioid cell type. Compared with retinal tissue, chromosomes 3,6,8 of tumor were detected by FISH. To use chromosomes 18 demonstrate repetitiveness of the experiment. Analyze the correlation of chromosome and pathology, tumor basal diameter, tumor thickness in choroidal melanoma. SPSS 15.0 was used for statistic analysis.Results There were 21 males and 15 females in the 36 patients who were between the age of 23 and 73, and all of them belonged to Han nationality. There were 16 left eyes and 20 right eyes. The average tumor basal diameter and tumor thickness were 12.5mm and 8.9mm. The eyes and ages in groups had no statistic differences.The pathology of 36 cases included 16 spindle cell type,10 mixed cell type,8 epithelioid cell type and 2 necrosis. The average tumor basal diameter of epithelioid cell type was larger than that of others, but it had no statistic differences. Tumor thickness of mixed cell type and epithelioid cell type was higher than that of spindle cell type, and it had statistic differences.2 cases were severe cellular necrosis, so they couldn't be detected the chromosome.34 cases included 13 loss of chromosome 3,9 loss of chromosome 8,7 loss of chromosome 6,4 loss of chromosome 3,8 and 4 loss of chromosome 3,6,8. No loss of chromosome 18 and no loss of chromosome in retinal tissue.13 cases of monosomy 3 included 3 spindle cell type,5 mixed cell type and 5 epithelioid cell type. 7 cases of monosomy 6 included 1 spindle cell types,3 mixed cell type and 3 epithelioid cell type.9 cases of monosomy 8 included 1 spindle cell type,4 mixed cell type and 4 epithelioid cell type.Loss of chromosome had no relation to tumor basal diameter and tumor thickness. Loss of chromosome had a strong relation to pathological classification. (P<0.05) Monosomy of mixed cell type and epithelioid cell type was more than that of spindle cell type. (P<0.05) Multiple loss of chromosome was more frequent in mixed cell type and epithelioid cell type. (P<0.05) Conclusion Loss of chromosome 3,6,8 existed in choroidal melanoma. Monosomy 3 is the most frequent, follow by loss of chromosome 6 and chromosome 8.Loss of chromosome had a strong relation to pathological classification. |
