Berberine, Evodia Drug Compatibility Study

Purpose:In this paper,basic on the theories of Chinese medicine,combined with advanced modern science and technology to study a comprehensive standardized research on the compatibility of Coptis chinensis Franch and Evodia rutaecarpa(Juss) Benth.Material and method:To the role of the chemical composition and efficacy, determining the best Preparated methods and combined ratio on the compatibility of Coptis chinensis Franch and Evodia rutaecarpa(Juss) Benth;combining on the method of Orthogonal Design and Uniform Design to selected the optimum extraction on this compatibility;The use of HPLC,to study the extraction of this compatibility on qualitative,quantitative,as well as pharmacokinetic.Results:1.According to the experimention,to determine the best preparation method of this compatibility is extract with alcohol together,the best combined ratio is 6:1.2.Using orthogonal design with the uniform design method to determine the best extraction process of this compatibility as follows:Coptis chinensis Franch(coarse) together with Evodia rutaecarpa(Juss) Benth jioned 8 times 70%ethanol to extract 2 times,2 hours every time.3.The use of HPLC,developed a fingerprint of the different proportion of this compatibility,the peak height ratio of Berberine and Evodiamine as an indicator in order to be able to distinction different combined ratio of this compatibility.4.Using the method of TLC,to identification the Jatrorrhizine,Palmatine Hydrochloride,Berberine Hydrochloride,Evodiamine and Rutaecarpine in this compatibility,the spots were clear,separation,Rf,repeatability and property of the method are professionals.5.The use of HPLC,to determine the cmponent indicators of this compatibility,its linear relationship was good,precision,stability,repeatability and recovery are in line with the standards.6.Using the method of internal standard,to study pharmacokinetic of Berberine in the extraction of the compatibility of Coptis chinensis Franch and Evodia rutaecarpa(Juss) Benth and Coptis chinensis Franch,the main pharmacokinetic parameters of the compatibility of Coptis chinensis Franch and Evodia rutaecarpa(Juss) Benth were:T_max(h)0.9000±0.6519,C_max(mg·L^-1)0.04740±0.01467,MRT_{(0～∞)}16.32±4.151; the main pharmacokinetic parameters of Coptis chinensis Franch were:T_max(h) 1.350±0.9287,C_max(mg·L^-1)0.02600±0.008631,MRT_{(0～∞)}16.71±2.355。Conclusion:To Determine the best preparation methods,combined ratio and the extraction process of the compatibility of Coptis chinensis Franch and Evodia rutaecarpa(Juss) Benth by the full norms research,set up a system of fingerprint and quality standard and improve the research of pharmacokinetics,the best combined ratio of this compatibility is consistent with the traditional Cuban Zuojin Pill(Huanglian,rutaecarpa 6:1),this result is as a proof to prove the reasonable of ancient combination and provide reference for further research and development of the compatibility of Traditional Chinese Medicine.
