Objective:To study the expression of endogenous hydrogen sulfide (H2S) in a rat APE model, and explore effect of exogenous administration of H2S donator NaHS and PPG on the rat APE model.Methods:1,The major divided group: To utilize eighty healthy wistar rats of Jiamusi university amimal experiment center, a half of female and male, the weight is from one hundred and seventy grime to two hundred grime.To be divided four groups in ran. Control group (C-group),model group (M-group),NaHS-treated group(S- group),PPG-treated group(P- group). 2,Make animal model: We constructed a rat APE model by injecting 3-4 emboli into the right jugular vei .3,Evaluation: To appraise the models doing well or not basis on the histomorphology and pathology of lung of rats .4,Target: All rats were sacrifieed on 1 hour,1 day and 3 days after treatmen respectively . 1 ml vascular blood was withdrawn from each animal to measure blood gas analysis and lung removed for histopathologic with microscope, the concentration of H2S in the plasma were detected; the activity of CSE in the lung tissue were measured by spectrophotometry.Results:1,In the C-group,the notable embolism in pulmonary arteries inflammatory cell infiltration around alveoli could be found by microscopy.2,PaO2 and PaCO2 were obviously lower at 1 hour after embolization (P<0.01), and PaCO2 was gradually increased to the normal.3,The contents of endogenous H2S in plasma and lung tissue were markedly reduced and the activities of H2S synthase in lung tissue were significantly were lower after embolization (P<0.05).4,NaHS-treated group rats showed emblism,hemorrhage,inflammatory reaction relative lighten,but blood gas analysis was not obviously improved.5,PPG-treated group rats were deteriorated. In the S-group,the plasma level of H2S, the activity of CSE in lung tissue were elevated than the M-group (P<0.05).But in the P-group,the plasma level of H2S, the activity of CSE were more decreased than the other groups (P<0.05). Conclusions:1,By injecting 3-4 emboli into the right jugular vei of the rats,successfully established an pulmonary fibrosis animal model. 2,Endogenous H2S may be involved in the generation and development of PTE.3,After exogenous administration of H2S donator NaHS can alleviate the formation of PTE. 4,After exogenous administration of PPG can aggravate the formation of PTE.
