Oviducts Ranae Antitussive The Chemical Composition Of The Active Site

Oviductus Ranae is the dry oviductus of the thely- Ranae temporaria chensinensis David,which is also called Ranae chensinensis oil.The medicine was recorded in Chinese pharmacopoeias from edition 1985 to edition 2005,it has the effectiveness of invigorating the spleen and stomach,nourishing the production of body fluid.Phamarcology studies indicated that Oviductus Ranae ethanol extraction has good antitussive effects,so this experiment carried out the studies to ethanol extraction chemical constituents.At first,oil-soluble impurities were extracted by SCCO₂, then five compounds were isolated from EtOAc fraction of 85% ethanol extraction by repeated c-18 silica gel ,sephadex LH-20 column chromatography and HPLC,LC-MS,GC-MS. The chemical structions of cholesta一5一en-3β,7β—diol (Hâ… ), 3β一hydroxy-cholesta-5一en一7一one (Hâ…¡)were fully determined by ¹H-NMR,¹³C-NMR and DEPT combining ESI-MS, the chemical structions of glycerin(Hâ…¢),alpha-Methyl-D-mannopyranoside(Hâ…£), cholesta-4,6-dien-3-ol (Hâ…¤) were analyzed by GC-MS because of light weight.Furthermore it is confirmed that cholesta一5一en-3β,7β—diol (Hâ… ), 3β一hydroxy-cholesta-5一en一7一one (Hâ…¡) were isolated from Oviductus Ranae for the first time.The paper studied the chemical constituents of Oviductus Ranae antitussive parts,which establishes foundation for further study of rare animal medicine Oviductus Ranae.
