Purpose: To study the effect of Astragalus extract to the skin of naturalaging mouse in total antioxidant capacity (T-AOC) and the ATP enzyme, and to observe the organizational structure of mouse skin before and after changes in drug use, to detect the function of Astragalus extract in delaying the role of natural aging in mouse skin.Material and method: 12 of female Kunming mice of 4-month-old as the youngcontrol group(A) , 60 of female Kunming mice of 18-month-old as the old age group, and then were randomly divided into 5 groups , with every group having 12 Kunming mice of 18-month-old : elderly control group(B), positive control group of older (C), low-dose group of Astragalus (D), middle dose group of Astragalus (E), high-dose group of Astragalus (F). All the mice were fed in the same natural environment. The mice of group A and group B were administered with 0.9% saline NS (20ml/kg); group D, group E and group F were administered with Astragalus extract of 4g/kg, 8g/kg, 16g/kg; group C were administered with suspension of vitamin E·C 3mg/kg. Operating 30 days, once a day. The first 31 days all the mice were executed and derived.Results:1. Compared with young control group (A), the total antioxidant capacity (T-AOC) and the ATP enzyme activity in skin of organizational structure of group B were significantly lower than group A, with significant differences (respectively was: p<0.01, p<0.01), descripting that with aging, T-AOC and ATP enzyme activity in the skin of natural aging mouse were reduced significantly; there is a significant difference of T-AOC in the skin of natural aging mouse of group D and E , compared with group A(respectively was: p<0.01, p<0.05); while group F and A had no significant statistical difference (p>0.05). The ATP enzyme activity in the organizational structure of skin in group C, D, E and F were significantly different, compared with group A (respectively was: p<0.05, p <0.01, p<0.01, p<0.05).2. Compared with groupB, theT-AOC were increased significantly in skin of organizational structure of group C, D, E and F(respectively was: p<0.01, p<0.05, p<0.05, p<0.01), descripting that suspension of vitamin E·C and Astragalus extract with different concentrations can increase the total antioxidant capacity(T-AOC), and prove the role of skin aging; the ATP enzyme activity in the organizational structure of skin in group C, D, E and F were significantly higher (respectively was: p<0.05, p<0.05, p<0.05, p<0.01), descripting that suspension of vitamin E·C and Astragalus extract with different concentrations can increase the enzyme activity of ATP of organizational structure in skin, to prove the role of anti-aging of Astragalus.3. In the group D, E and F , the content of the total antioxidant capacity (T-AOC) and the ATP enzyme activity in group F was highest, followed by group E, and group D at least content. In the total antioxidant capacity (T-AOC) of mice skin tissue, group D and group F had a significant statistical difference (p<0.01); while group D and E,group E and F, all had no significant statistical difference (respectively was: p>0.05, p>0.05). In the ATP enzyme activity, group F and E,group F and D all had significant statistical difference (respectively was: p<0.05, p<0.01); while group D and E had no significant statistical difference (p>0.05).4. Morphological observations indicate that: compared with the young control group (A), the structure changes of epidermal and dermal elderly control group (B) were in state of aging; but in the positive control group (C), low, middle and high - dose group of Astragalus extract (D, E, F) , the structure of epidermis and dermis were better than the elderly control group (B), had not yet achieved the state of the skin structure of young control group (A).Conclusion:1. The total antioxidant capacity(T-AOC) and the ATP enzyme activity in skin tissue of natural aging mouse decrease significantly.2. Astragalus extract can improve the total antioxidant capacity(T-AOC), increase ATP enzyme activity, slow down skin aging. 3. In high concentration , the role of Astragalus extract that increasing the total antioxidant capacity (T-AOC) was stronger than low-dose of Astragaloside extract. The role of Astragalus extract that increasing the enzyme activity of ATP was the strongest among the three groups of different dose of Astragaloside extract.4. From the morphological observation: the control group compared with the young control group(A), the skin structure of natural aging in mice were shown, and the Astragalus extract can slow down the state of aging effectively.
