Objectives:Hyoscyamus niger L.,which originally called "LangDangzi",is the dry and mature seed of henbane plant,it is hypertoxic and has the effect of relieving spasm and pain,relieving uneasiness and fixed pain.its main function are curing gastrospasm, dyspnea with coughing,vecordia,anti-shock and improving microcirculation,in recent years,we have further found its multilateral pharmacological effects and clinical functions.Chinese pharmacopoeia(2005) had recorded Hyoscyamus niger L.but no processing product,however,the ancient documents all recoreded many processing methods,in order to perfect Hyoscyamus niger L.'s quality standard,it is necessary to do the further study of its processing technique so that we can find a proper one that may minus poison and have synergic effect,be steady,controllable,easy to popularize and fit for mass production,based on a great number of reference documents,my thesis studies the prcessing technique of Hyoscyamus niger L.,selects the best processing parameter in order to lay a fundation for making its quality standard,and more rationally and more effectively provide scientific reference for clinical meditation.I'll do the research from the following three points.Material and method:The chemical components' research of Hyoscyamus niger L.,Adopting hyphenated technique GS-MS,HPLC(high performance liquid chromatography)ultraviolet to analyse and compare every components of Hyoscyamus niger L.that before processing and after processing:Studies on pharmacological action of Hyoscyamus niger L.'s raw product and processing product,through the hot plate test and writhig method on the rats to watch the analgesic effect,in the experiment of using xylol to make the rats' auricle swelling,watching the anti-infalmmatory effect,at last, according to orthogonal test table to determined the best technique。Results:1.The chemical components' comparison and research of Hyoscyamus niger L.'s raw product and processing product.Adopting hyphenated technique GS-MS to analyse the volatile oil components of Hyoscyamus niger L.that before processing and after processing,respectively got 29 compounds,compare to the raw product,I found that most of the components' contents of sole frying product decreased,palmitate decreased by 1.4486,oleate decreased by 1.2698,linoleic acid obviously decreased by 2.6799;while the contents of acetaldehyde increased by 1.3414.compare with vinegar products and raw products,I found that except minor components'contents such as 1-Pentanol(decreased by 0.0083), 2-Nonanone(decreased by 0.0038),6-Undecanone(decreased by 0.0296),2-Undecanone(decreased by 0.0305),2-Octenal,2-butyl(decreased by 0.0521) decreased,all the other greatly increased,the most obvious was linoleic acid,which increased 21.0523,as much as 6 times.By the comparison of GC,chromatography,I found that the component of fatty acid in Hyoscyamus niger L.and its processing product;adopting an internal standard to calculate the contents,I found that the content of fatty acid chemical components in the raw product and processing product was almostly unchanged;through using hyphenated technique GC-MS to analyse the components of fatty acid in Hyoscyamus niger L.,the result showed that the 11 kind of fatty acids take 99.2%of the tatal fatty acid,the content of linoleic acid reached to 66.5%,oleate was 17.26%,which took the second position.Adopting HPLC method to compare the cotents of scopolamine hydrobromide, atropine sulfate,racanisodamine in Hyoscyamus niger L.that before processing and after processing,the result told that sole frying can make the contents of scopolamine hydrobromide,atropine sulfate,racanisodamine in Hyoscyamus niger L.respectively decrease(mg·g-1) 0.0187,0.0202,0.000602;while those contents in the vinegar processing Hyoscyamus niger L.respectively increased(mg·g-1) 0.0386,0.0446, 0.000748.By the comparison and research of water decocting and water immersion to extract polysaccharide,the percent of polysaccharide extration in using water decocting method is 0.05%lower than in using water immersion method;from analysing the orthogonal experimental results of extraction in using water immersion method,the best technique of Hyoscyamus niger L.'s crude polysaccharide extraction is A2B3C3D1,that is using 8 times amount of water,the temperature controlled in 100℃,extract 3 times,2 hours per time.Using ultraviolet spectrometry to determine the contents of polysaccharide in Hyoscyamus niger L.'s raw product,sole frying product and vinegar product,the result respectively are 21.93 mg·g-1,20.35 mg·g-1,20.51 mg·g-1,showing the processing affects little on polysaccharide.2.Studies on pharmacological action of Hyoscyamus niger L.'s raw product and processing product.At first,I do an acute toxic experiment on Hyoscyamus niger L.'s raw product and processing product.adopting maximum tolerance dose method,using the water-decocted liquid in maximal dose of the raw product and processing product to do gastric perfusion on rats expendably,I found all the rats didin't die.the maximun tolerance dose of the mouse drinking the water-decocted liquid of the raw product and processing product are 200g·kg-1,190 g·kg-1 and 192 g·kg-1,respectively equal to 8333, 7917 and 8000 times clinical dosage.Secondly,through the hot plate test and writhig method on the rats to watch the analgesic effect,I got that the Hyoscyanmus niger L.'s raw product and its two processing products have great analgesic function,moreover the analgesic effect has no obvious difference between them.Finally,in the experiment of using xylol to make the rats' auricle swelling, watching the anti-infalmmatory effect,the result showed that the positive control group, raw product group,sole frying product group and vinegar processing product group,all have obvious inhibition effect for the swelling of auricle in rats caused by xylol.that is to say Hyoscyamus niger L.and its two processing products have well inhibition effect. and through the swelling inhabition percent we can get that the vinegar processing product's inhibition effect is stronger than the sole frying product's;while the sole frying products's is stronger than the raw product's.3.Studies on processing techniqueSole frying method experiment result showed,the contents of scopolamine hydrobromide,atropine sulfate gradually decreased with the temperature of sole frying's increased and the time's passing,it proved that the over hot temperature will destroy alkaloid,making alkaloid decrease,however,in the vinegar processing mathod, by observing the drying method,the amount of vinegar,stuffining and moistening time, microwave time and microwave strength,making sure the factor level,arranging test according to orthogonal test table,taking the contents of scopolamine hydrobromide and atropine sulfate as indicator,I do the optimal selection of Hyoscyamus niger L.'s best vinegar processing technique,determined the best technique was to plus rice vinegar 40kg per bupleurum medical slices 100kg,stuffin and moisten 2 hours on low-power microwave,and then dry 4 minutes.Conclusion:In aspect of the chemical components' research of Hyoscyamus niger L. that before processing and after processing,it is a definity changing for the volatile oil and fatty acid,while it is have little change in polysaccharide and fatty acid;studies on pharmacological action of Hyoscyamus niger L.'s raw product and processing product. the Hyoscyamus niger L.that before processing and after processing have a small gas chronatography for rat,and have a definite of analgesic effect,moreover the analgesic effect has no obvious difference between them.however,through the swelling inhabition percent we can get that the vinegar processing product's inhibition effect is stronger than the sole frying product's;while the sole frying products's is stronger than the raw product's,at last,on the base of the chemical components' research and pharmacological action,I do the optimal selection of Hyoscyamus niger L.'s best vinegar processing technique,determined the best technique.
