| Refined Qingkailing injection(RQKL) is composed of bile powder,baicalin, honeysuckle,gardenia,etc.Effectiveness:antipyretic and Dehumidity,cooling blood and detoxification,diluting phlegm and eliminating stagnation.It would be used for the treatment of upper respiratory tract infection and acute lung infection,etc.Upper respiratory tract infection and acute lung infection should be in the area of Warm-disease of traditional Chinese medicine caused by Warm evil with emphasis on heat,Ying harmed easily being dryness,such as a class of acute exogenous fever. Chinese medicine believes that the weak ferrite and also with infection of Six evil elements or Pestilence is the major pathogeny.The main pathological process is changes from appearance to inside.Modern medicine believes the upper respiratory tract infection is caused of a wide range of pathogenic bacteria or viruses infection and mainly infract the lung.The main characteristics of the disease is the influence lung function,high fever,even coma worse.Based on traditional Chinese medicine theory of warm disease awareness and the pathogenesis of modern medicine,the series investigations were carried out about the effect of RQKL on:the anti-SARS virus in vitro;endotoxin-induced fever in rabbits; rabbit viral pneumonia;endotoxin-induced multiple organ injury in rats;bleomycin induced lung injury in rats.Grouping and dosage:Dose in rats as follows:RQKL high-dose group:2.78g crude drug / kg,middle dose group:1.39 g crude drug / kg,low-dose group:0.70 g crude drug / kg;respectively, equivalent to clinical dose as 16,8 and 4 times.Dose in rabbits as follows:RQKL high-dose group:1.20g crude drug / kg,middle dose group:0.60 g crude drug / kg, low-dose group:0.30 g crude drug / kg;,respectively,equivalent to clinical dose as 6.9,3.5 and 1.7 times.Control and model control:Water. Results:RQKL has a certain effect on anti-SARS virus,CPE method,its prevention, suppression and inactivation of drug half effective concentration(IC50) to SARS-COV-Pll,SARS-COV-8 as follows:32.5mg / ml,65.0mg / ml and 32.5mg; the minimum effective concentration(MIC) as follows:16.3mg / ml,32.5mg/ml and 16.3mg/ml;the therapeutic index(TI):4,2 and 4,respectively.The endotoxin-induced temperature rise in rabbits can be significantly inhibited by RQKL.Not only the elevated body temperature reduced significantly,the animal body temperature response index decreased also.RQKL has a certain inhibition to the temperature increase in viral pneumonia rabbits.Endotoxin poisoning condition is the deterioration of the upper respiratory tract infection.The models were induced by injection of endotoxin into the rat.Result:the anti-endotoxin Effect of RQKL is definite.The injures in rat lung,heart,kidney,and liver were evidently recoveried by treatment with RQKL At the same time,RQKL can increase the serum concentration of SOD and reduce the concentration of MTA. The rabbit viral pneumonia models were induced by injection of Sendai virus into the rabbit trachea directly.Experimental result revealed the inflammation injury in rabbit lung bronchus and pulmonary alveoli were inhibited by RQKL,including alveolus inflammatory cell infiltration,cell infiltration bronchiolitis,erythrocyte sedimentation, interstitial inflammatory cell infiltration and widening,etc.The temperature rise of rabbits of treated groups were much lower than control group.The rat lung injury models were induced by injection of BLM into the rat trachea directly.Result revealed the inflammation injure in rat lung bronchus and pulmonary alveoli,and pulmonary fibrosis in pulmonary mesenchyme were inhibited by the treatment of RQKL. Conclusion:The refined Qingkailing injection has some antiviral activity;better antipyretic effect;obvious impact of anti-endotoxin;significant protective effect on viral and chemical-induced lung injury. |
