Chemical Composition. The Liuwei Rehmaionoside Sugar Glycosidic Parts Research

Liuwei Dihuang Gantang, a new crude drug of classⅤ, was composed of glycosides, oligosaccharides and polysaccharides. It was researched and developed by Institute of Pharmacology and Toxicology, Academy of Millitary Medical Sciences and was submitted to the Center of Drug Evaluation of State Food and Drug Administration. Liuwei Dihuang Gantang was modified from Liuwei Dihuang Decoction based on a lot of basic research. Among these three fractions, the glycosides fraction was confirmed to be the main immunomodulation and endocrine regulation constituent, but its chemical composition was still not clarified fully.A systematic research on the chemical constituents of the glucosides was carried out based on the large-scale preparation of the fraction, and the preparation was processed rigorously according to its preparation technology. Eleven compounds were isolated from the glucosides by column chromatographies on silica gel, Sephadex LH-20, RP C-18, preparative TLC and semi-preparative HPLC. Nine iridoid glycosides, one pinane monoterpene glycoside and one phenolic glycoside were isolated and identified as 7-dehydrologanin(1),7α-O-methylmorroniside(2),7β-O-methylmorroniside(3),7α-O-ethylmorroniside(4),7β-O-ethylmorroniside(5),morroniside(6),sweroside(7),loganin(8),paeoniflorin(9),paeonolide(10),loganic acid(11). All the above compounds were isolated from Liuwei Dihuang Gantang for the first time and they laid a foundation for the better quality control of the glycosides fraction .
