| Fructus Forsythiae is the fruit from plants of Fosrythia suspensa (Thunb.) Vahl, classified unripe Fructus Forsythiae and ripe Fructus Forsythiae. Fructus Forsythiae has all kinds of pharmacological activity consisting of anti-bacteria and anti-virus, antipyretic and anti-vomitting, antihypertensive and diuretics, resisting hepatic injury, antiendotoxin et al. It is often used to treat Cold , Scrofula, Urinary infection etc. As one of the traditional Chinese clinical herbs, Fructus Forsythiae has a relatively high medicinal value. It mainly contains terpenes, phenylethanol, flavonoid, lignan, alcohol, ester, aldehyde, ketone, volatice oil and so forth. Phillyrin, phillygenin, oleanolid acid, rutin, forsythoside are the main effective compositions among them.The dissertation consists of four chapters focusing on the study of extraction method and metabolism of the effective ingredient of Fructus Forsythiae by intestinal bacteria. The contents are as follows:Extraction and separation of effective compositions from Fructus Forsythiae were studied by using the Macroporous adsorptive resins and preparative high performance liquid chromatography. 2 compounds were isolated from Fructus Forsythiae. On the basis of analysis of UV, IR, NMR, MS data and measuring of physico-chemical characters, the structures of 2 compounds were elucidated as forsythoside( I ), Rutin( II). The results show that the Macroporous adsorptive resins D101 enriched the effective compositions, at the same time, could remove a lot of extraneous compounds. Preparative high performance liquid chromatography has many advantages over traditional chromatographic methods such as high efficiency, high loading capacity and high recovery.To study and compare the metabolism of phillyrin and rutin in vitro intestinal flora from rat and rabbit. Intestinal flora of rat and rabbit were anaerobically cultured at 37℃in vitro. The main metabolites of phillyrin and rutin were separated and quantified by HPLC-MS. Phillyrin and rutin was quickly metabolized by rabbit intestinal flora, and gently by rat intesinal floral . In vitro, Phillyrin and rutin are metabolized by intestinal flora of rat and rabbit.
