Matrine On S <sub> 180 </ Sub> Sarcoma Mouse Tumor Blood Vessel Formation Inhibition Studies

Objective:To investigate the effects of matrine on the expression of vascular endothelial growth factor(VEGF) and microvascular density(MVD) in S180 sarcoma in miceMethod : The model for transplanted S180 fluid tumor in mice was set up . 40 Balb/c mouse,18-22g,were separated to 4 group randomly according to mass (n=10 each ), S180 sarcoma cell were implanted into all groups by subcutaneous injection. After 48h, the 4 groups mice were given 0.9%NaCl 0.2mL, matrine(50mg/kg),CTX(20mg/kg),matrine(50mg/kg)+ CTX(20mg/kg).After the drug was given continuously for 15 days, the mice were killed by decapitation, caculate tumor inhibiting rates by tumor weight .Observe matrine's inhibition of the S180 sarcoma in mice from developing by pathological diagnosis in vivo ascitic ; observed the expression of VEGF protein by SABC immunohistochemistry method and apoptosis by electron microscope.Result:1. The mice of control group had tarnished fur,samll amout of food , spirit sagged , moved targily,the weight increase significancely was less than other groups,the general state of mice in matrine+CTX group was best;2.Under light microscope,the nascent capillary in group Control group was more dense than group matrine,group CTX and group matrine+CTX;3.Under electron microscope , the wall of the nascent capillary in the tumor was flimsy ,It was the pseudopod of endodermis cell which contacted endodermis cells, the dense between endodermis cells was wider than normal, the basement membrane was thin and relax.4.VEGF protein was mainly stained in cytoplasm,nuclear membrane with brown color,theâ…§factor was mainly stained in cytoplasm,nuclear membrane with brown color,too. The expression of VEGF and the MVD in group matrine, group CTX and group matrine+CTX was significantly decreased as compared with that in group control .Conclusion: 1. Matrine can raise the life quality of S180 sarcoma mice,depressthe side-effect of western medicine .2. Matrine can inhibit the formation of the new blood vessels in the tumor of S180 sarcoma mice. 3. Matrine can inhibit the expression of VEGF in the tumor of S180 sarcoma mice. 4. Matrine can decrease MVD in the tumor of S180 sarcoma mice. 5.Through inhibiting the formation of the new blood vessels in the tumor of S180 sarcoma mice, decreasing MVD in the tumor and inhibiting the expression of VEGF in the tumor, matrine can inhibit metastasis of tumor.6. Through inhibiting the formation of the new blood vessels in the tumor of S180 sarcoma mice, decreasing MVD in the tumor and inhibiting the expression of VEGF in the tumor , Matrine can raise the life quartily and inhibit the formation of the new blood vessels in the tumor. 7. Matrine have co-operation with cisplatin.
