Huanglian Of Aconite Attenuated Synergistic Compatibility Preliminary Study | | Posted on:2009-08-24 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y Zhang | Full Text:PDF | | GTID:2204360272964438 | Subject:Pharmacy | | Abstract/Summary: | PDF Full Text Request | | Purpose:Discuss whether or not Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata have compatibility relation reducing toxicity and original(increasing) effect.Method:①Adopt Bliss method to detect acute toxicity of Rhizoma Coptidis,Radix Aconiti Lateralis Praeparata and their cooperative group.②Adopt spectrophotography to detet their influence on liver function(ALT,AST) and kidney function(BUN) in mice blood serum.③Adopt semi-solid paste filling taken method to detect their influence to gastric residue rate and small intestine advancing rate.④Measure their influence to cardiac muscle physiology and eleetrogastro-physiology by BL-420E+ four path physiologic recorder.⑤Review their influence on lapactic mice model by adopting Radix et Rhizoma Rhei mice model.Result:①LD50 credibility interval of Rhizoma Coptidis is 3.65 to 5.53g/kg.Mice death rate of Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:1 is obviously lower than single Rhizoma Coptidis group(P<0.05).②Rhizoma Coptidis group,Radix Aconiti Lateralis Praeparata group and Rhizoma Copfidis and Radix Aeoniti Lateralis Praeparata 1:1 and 1:2 cooperative group rise liver function ALT value of mice blood serum obviously(P<0.01).Rhizoma Coptidis group can rise AST value obviously and AST value of the Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:2 is obviously lower than that of the Rhizoma Coptidis group(P<0.01). Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:1 can reduce BUN of mice blood serum obviously(P<0.01).③Each administrating group can inhibit gastric emptying motion and small intestine promoting motion obviously(P<0.01). Inhibition of Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:1 and 1:2 to stomach and intestines is more obvious than single aeontum fischeri group P<0.01; Inhibition of Rhizoma Copfidis and Radix Aeoniti Lateralis Praeparata 1:2 to gastric emptying is more obvious than single Rhizoma Coptidis and Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:1.Each administrating group has not obviously influence to gastric electric slow wave frequency and swing of rat but results in gastric electric slow wave rhythm disorder.Rhizoma Coptidis and Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata(1:2) are more obvious(P<0.01).④Rhizoma Coptidis group,Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:1 and 1:2 cooperative group can improve diarrhea obviously preferential than Radix Aconiti Lateralis Praeparata.⑤Rhizoma Coptidis group can reduces heart rate of rate obviously for administrating 45min and 60min and electrocardio P waves are reversed obviously(P<0.01) at each time period;Radix Aconiti Lateralis Praeparata group tends to induce reversed P wave but no statistic meanings;cooperative group has not obvious influence to electrocardio.Conclusion:①The LD50 of Rhizoma Coptidis is 4.65±0.94g/kg and can causing liver damage;it can inhibit gastrointestinal motility having strong anti-diarrhea effect;it also can intervene gastric and cardiogram.②Radix Aconiti Lateralis Praeparata has a certain acute toxicity having influence on liver toxicity;it inhibits gastroenteritic motion in a certain degree;it has no anti-diarrhea effect induced by rhubarb.③Rhizoma Coptidis and Radix Aconiti Lateralis Praeparata 1:1 cooperative group can reduce acute toxicity of the Rhizoma Coptidis obviously.
