| In recent years,the ulcerous colortitis(UC) incidence rises continuously,which draws more attentions from medicine vocation.Chinese medicine has better curative effects and less adverse reactions,but the laggard dosage forms restrict its function.Thus the research on oral target preparations for colonitis in TCM is very important and urgent.According to Chinese medicine theory and pharmacological trials,the innovative compounded Chinese medication Jiechangkang for UC was sifted systematically.The prescriptionis composed of astrageloside,amylase,ma-trine,oxymatrine.Established internal standards for astrageloside and amylose extractions that have no national standards.At those effective components above,developed reaserches on the molding technics of two-direction type stomach-colon target preparations of Jiechangkang.As to patent drug,established a TLC method for identifying astragaloside,matrine and oxymatrine as well as a HPLC method for determination of matrine and oxymatrine, formulated the content limits of matrine and oxymatrine in preparations.Regarding the accumulative dissolution percentage of matrine and oxymatrine as assessing indexes, established a method of in vitro dissolution determination for preparations. Besides,investigated some factors such as rev of PH value of solvents and tablets'hydrosopic property that could affect preparations" release.Experiments indicate that the inner-outer structural type is much better than above-under structural type in the aspects of release and stability.Reviewed theinner-outer structural type stomach-colon target preparation in vivo release by giving an enema,as a result,it could achieve the objective of colon oriented release in a normal animal body.This research discussed new thoughts about Chinese medicine system made of two or more ingredients.The conclusion can offer some advice and experience for the studies and developments of oral colon target preparations of Chinese medicine in the future. |
