| Objective:Glycyrrhizae belongs to Glycyrrhiza of pulse family plant,it's root and stem are the traditional Chinese medicine that be commonly used.It has long-term medication history,in civil of our country.The materia medical of successive dynasties all record it and it is the one of common medicine of clinical tradition Chinese medicine.70 prescriptions make use of glycyrrhizae in 113 prescriptions of Treatise on Febrile Diseases and it is frequently used.It can result in extensive influence of organism and there is therapeutic effect invariably in many diseases.Fired Glycyrrhizae decoction comes from Treatise on Febrile Diseases of Zhang Zhongjing.It is a classical famous decoction and it can cure deficiency of Heart-Qi,deficiency of Heart-Blood,regularly intermittent pulse,palpitation and so on.It is used to cure cardiac arrhythmia in Modern clinical and gets good therapeutic effect.In recent years,research discovers that fired glycyrrhizae decoction can improve immunity and resist aging.But it is not supported that cure deficiency of Heart-Qi and deficiency of Heart-Blood in blood generate and hematopoietic regulation field.At the same time,it can't be widely used in the clinical application because of lake of related experiment data.In the present study,our aim is to explore the effect of fired glycyrrhizae decoction on hematopoietic regulation and its mechanisms and provide the experimental data for the clinic use and exploitation.Methods:In the present study,the model of myelosuppressed mice is prepared by 60Co irradiation and cytotoxic chemotherapeutic compounds(cyclophosphamide). Peripheral blood cell count,myeloid tissue section,hemopoietic progenitor cell culture,immunohistochemistry and flow cytometry were used to analyze the survival and proliferation of HSCs,cell cycle of bone marrow nucleated cells and the expressions of apoptosis-related protein,respectively.We explored systematically and deeply the effect of Fired Glycyrrhizae decoction on hematopoietic system and its mechanisms on cell,protein in myelosuppressed mice.Results:Within the range of certain dosage or concentration: (1) Fired Glycyrrhizae decoction could promote obviously the increase of the number of bone marrow cell(BMC),peripheral blood leucocyte number,peripheral blood erythrocyte number,hemoglobin amount and the area of hematopoietic tissue in a dose-dependent manner in myelosuppressed mice.Along with the prolongation of time,the number of BMC and the area of hematopoietic tissue also increase gradually,but it dose not display evident promotion on blood platelet.(2) Fired Glycyrrhizae decoction could promote the colony formation of burst-forming unit-erythroid(BFU-E),colony-forming unit-erythroid(CFU-E) and colony-forming unit-granulocyte macrophage(CFU-GM),but each colony showed various features.(3) Fired Glycyrrhizae decoction could promote the entry of BMC into cell cycle, and decrease the cell proportion of G0/G1 phase,and increase the cell proportion of S phase and G2/M phase in myelosuppressed mice.At the same time,it could also result in the increase of proliferation index(PI) of BMC.(4) Fired Glycyrrhizae decoction could promote significantly the expression of Bcl-2 protein,inhibit the expression of Bax protein of BMC in myelosuppressed mice.Conclusions:Fired Glycyrrhizae decoction could promote the generation and differentiation of hematopoietic progenitor cell,and promote the recovery of peripheral blood and bone marrow in myelosuppressed mice,it could also promote the recovery of hematopoietic function by accelerating G0/G1-S phase and S-G2/M phase transition,promoting the protein expression of Bcl-2,inhibiting the protein expression of Bax in myelosuppressed mice. |
