| The dried and ripe fruit or leaf of Illicium macranthum A.C.Smith,which belongs to Illiciaceae is a folk Chinese herbal drug.It is acid, hot, extremely poisonous. It promotes tissuw regeneration for treament of ulcer and scabies. It is also a insecidal. The chemical constituents of the most species of Illiciaceae have been studied, however there are no enough chemical data for the fruits of Ilticium macranthum. The thesis deals with the chemical constituents of Illicium macranthum and bioactivity of sesquiterpene lactones from Illicium macranthum. This thesis is composed of two sections.Section one:Literature Review1.Introduce the studies on chemical constituents of Illiciaceae;2.Introduce the pharmacological action of Illiciaceae.Section two:Experiment Studies1.Chemical constituents of the pericarps of Illicium macranthum. The methanol extract was devided into four fractions by silica chromatography eluting with petroleum,dichloromethane,dichloromethane:EAc(1:1) and methanol. This paper is dedicated to the seperation of dichloromethane:EAc(1:1) part.As results, twelve compounds were isolated from the pericarps of Illicium macranthum.Eleven of them were identified on the basis of chemical and spectral data, including MS,1H-NMR,13C-NMR,NOSEY et al., the structure of the compounds were elucidated as 6-deoxypseudoanisatin(1),pseudoanisatin (2),protocatecheuic acid(3),2-oxo-6-dehydroxyneomajucin(4),β-sitosterol(5),6-deoxyneomajucin(7),pseudomajucin(8),anisatin(9),shikimic acid methylester(10),shikimic acid(11) and daucosterol(12).All the compounds were first isolated from the species.Among them,2-oxo-6-dehydroxyneomajucin(4) and 6-deoxyneomajucin(7) are new compound, the rest compounds are known. So we get the conclusion of sesquiterpene lactones and organic acids are the main constituents of the pericarp of lllicium macranthum.2.The anti-inflammatory effect and analgesic effect of the sesquiterpenes from Illicium macranthum. Through the acetic acid-induced writhing test in mice to study the analgesic effect, the result showed that the extraction of the pericarps of Illicium macranthum can significantly reduce the writhing times of the mice at the dosage of 16.3mg/kg;and through the xylene-induced ear edema in mice to study the anti-inflammatory effect, the result showed that the extraction under the dosage of 17.1mg/kg can't inhibit ear inflammation induced by xylene in mice.So we think the extraction has analgesic effect and no anti-inflammatory effect. This paper introduces the studies on chemical constituents and the pharmacological effects of sesquiterpenes from the pericarps of Illicium macranthum for the first time, providing determinate evidence for better applying and developing the species. |
