Objective: (1)The aim of this study was to investigate the expression of Livin and bcl-2 protein in colorectal carcinoma,adenomas and normal tissues, to explore the relationship between Livin expression and bcl-2 expression. (2) By analyzing the expression of Livin and bcl-2 and colorectal cancer clinical pathological parameters, the aim was to explore the degree of malignancy and the possibility of progress of the colorectal carcinoma.(3) By analyzing the role of Lycium barbarum polysaccharide inhibition colorectal carcinoma cell, which was to explore Lycium barbarum polysaccharide inhibit the expression of Livin and bcl-2 protein in colorectal cancer cell .Methods:(1) The protein expression of Livin and bcl-2 were detected in colorectal carcinoma,adenomas and normal tissues respectively by Immunohistochemistry.(2)The role of Lycium barbarum polysaccharide inhibition colorectal cancer cell was analysed by MTT assay .(3) The apoptosis of tumor cells was detected by DNA- Ladder assey . (4) The changes of the protein expression of Livin and bcl-2 in sw480 cell by the intervention of Lycium barbarum polysaccharide were detected by Western-blotting assay .Results:(1)Livin protein was detected in 17 of 34 colorectal carcinoma cases (50.00%), but No Livin protein was detected in adenomas and normal colorectal tissues. The protein expression of bcl-2 were detected in colorectal carcinoma,adenomas and normal tissues respectively was 58.82%(20/34),80.00%(8/10)and 12.50%(1/8). The protein expression of Livin of colorectal carcinoma was significant correlation with colorectal adenomas and colorectal normal tissues(P﹤0.05). Bcl-2 protein is highly expressed in colorectal carcinoma and adenomas tissues, The protein expression of Bcl-2 was significant correlation between colorectal carcinoma and normal tissues(P﹤0.05). There was no significant correlation of the protein expression of Bcl-2 between colorectal carcinoma and normal tissues(P﹥0.05)。(2)There was no significant correlation between the protein expression of Livin and age,sex,Dukes stage,lymph node metastasis(P﹥0.05), but the protein expression of Livin was closely related to infiltration degree of carcinoma tissues and degree of tumor differentiation (P﹤0.05). There was no significant correlation between Bcl-2 gene expression and age,sex,degree of tumor differentiation,Dukes stage,lymph node metastasis(P﹥0.05), but the expression of Bcl-2 protein was closely related to infiltration degree of carcinoma tissues(P﹤0.05).(3) In addition to 100 ug / ml and 100 ug / ml (P> 0.05), there was significantly different between the control group and the other groups (P <0.05).(4) The strip of cell apoptosis was found by DNA-Ladder. (5) The protein expression of the drug groups was decreased significantly than the control group was detected by Western-blotting assay .Conclusion:①Livin gene is highly expressed in colorectal carcinoma tissues, but shows without expression in colorectal adenomas and normal tissues . There was no significant correlation between Livin gene expression and age,sex,Dukes stage,lymph node metastasis, but the expression of Livin gene was closely related to infiltration degree of carcinoma tissues and degree of tumor differentiation.②Bcl-2 gene is highly expressed in colorectal carcinoma and adenomas tissues, but shows lowly expression in colorectal normal tissues .There was no significant correlation between Bcl-2 gene expression and age,sex,degree of tumor differentiation,Dukes stage,lymph node metastasis, but the expression of Bcl-2 gene was closely related to infiltration degree of carcinoma tissues .③The expression of Livin protein was positively related to the expression of Bcl-2 protein in colorectal carcinoma tissues.④The expression of Livin and bcl-2 protion in colorectal carcinoma tissues had correlation with tumor cell proliferation.⑤Lycium barbarum polysaccharide can inhibit the growth of tumor cells.⑥The expression of Livin and bcl-2 protion in colorectal carcinoma tissues was inhibited by Lycium barbarum polysaccharide ,which induced tumor cells to apoptosis.
