AIM:TO probe the expression of matrix metalloproteinase-9(MMP-9),carcinoma suppressor gene PTEN in colorectal carcinoma,colorectal adenoma and normal mucosa of large intestine and their correlation with the clinical pathological factors in colorectal carcinoma,further investigate the relationship between them in colorectal carcinoma.METHODS:EliVision immunohistochemical method was to examined the expression of MMP-9 and PTEN in colorectal carcinoma,colorectal adenoma and normal mucosa of large intestine ,analyze their correlation with the clinical pathological factors in colorectal carcinoma and the relationship between expressions of them in colorectal carcinoma.RESULTS: 1.The expression of MMP-9 detected in colorectal carcinoma was significantly higher than that in normal mucosa of large intestine (56.5% vs 15%, P<0.01) . The expression of MMP-9 detected in adenoma was significantly higher than that in normal mucosa of large intestine (45% vs 15%, P<0.05). The expression of MMP-9 detected in carcinoma was not correlated with that in adenoma (56.5% vs45% P>0.05).The expression of PTEN detected in colorectal carcinoma was significantly lower than that in normal mucosa of large intestine (39.1% vs 95% ,P<0.01). The expression of PTEN detected in adenoma was significantly lower than that in normal mucosa of large intestine (65%vs95%, P<0.05). The positive rate of PTEN detected in carcinoma was not correlated with that in adenoma (39.1% vs65% ,P>0.05). 2.The expression of MMP-9 was negatively correlated with histological class (P<0.05).It was positively correlated with lymph node metastasis(P<0.05) and the depth of invasion(P<0.05). The expression of PTEN was positively correlated with histological class (P<0.05).The expression of PTEN was negatively correlated with lymph node metastasis(P<0.05)and the depth of invasion(P<0.05). The expression of MMP-9 and PTEN had no correlation with the tumor size as well as the age of patients.3.There existed significant correlations between the expression of MMP-9 and PTEN in colorectal carcinoma(r=-0.797,P<0.05).CONCLUSIONS:1.The expressions of MMP-9 in colorectal carcinoma and adenoma were significantly higher than that in normal colorectal tissues,the expressions of PTEN in colorectal carcinoma and adenoma were significantly lower than that in normal colorectal tissues , the high expression of MMP-9 and the low expression of PTEN could help the diagnosis of colorectal carcinoma at early stage.2.The expression of MMP-9 and PTEN significantly correlated with biological factors and they were one of important markes that determining molecular and biological feactures of colorectal carcinoma. 3.There existed significant correlations between the expressions of MMP-9 and PTEN in colorectal carcinoma.PTEN can regulate the expression of MMP-9 by many ways to affect the pathogenesis,development and metastasis of colorectal carcinoma.
