Part 1:Trichostatin A inhibited growth of human anaplastic thyroid carcinoma cells DRO in vitroObjectives:To investigate the effects of Trichostatin A on the growth of human anaplastic thyroid carcinoma cell line DRO in vitro.Methods:anaplastic thyroid carcinoma cell line DRO was treated with 0,0.1μmol/L,0.2μmol/L,0.5μmol/L,1.0μmol/L,1.5μmol/L Trichostatin A and observed at different time.The cell viabilities were measured through methyl thiazolyl tetrazolium.Flow cytometry was used to examine apoptosis and cell cycle when anaplastic thyroid carcinoma cell line DRO was incubated with 0 or 1.0μmol/L TSA for 24,48,72h.Results:treated with 0.1μmol/L,0.2μmol/L,0.5μmol/L,1.0μmol/L and 1.5μmol/L TSA,the proliferation of DRO cells was significantly inhibited.IPCM showed that there were obvious morphologic changes after TSA treatment,refraction rate of the cells under experiment decreased,cells turned round and floated round with the cell membrane shrinking.After treated by 1.0μmol/L TSA for 48 h,the proportion of cells in G2/M phase increased significantly compared with control group.Conclusion:TSA significantly inhibits growth of the human anaplastic thyroid carcinoma cells DRO in vitro. Part 2:Trichostatin A change the expression of hTERT -mRNA and p21waf1-mRNAObjectives:To investigate the effects of Trichostatin A on the expression of hTERT-mRNA and p21Waf1-mRNA in human anaplastic thyroid carcinoma cell line DRO in vitro.Methods:human anaplastic thyroid carcinoma cell line DRO was incubated with 0,0.1μmol/L,0.2μmol/L,0.5μmol/L,1.0μmol/L,1.5μmol/L TSA and the expression of hTERT-mRNA and p21waf1-mRNA was evaluated by RT-PCR at 24,48,72 h.Results:the hTERT-mRNA expression was down-regulated at 48 h but p21waf1-mRNA did not change significantly.Conclusion:Trichostatin A inhibits growth of anaplastic thyroid Carcinoma by down-regulated the expression of hTERT-mRNA.
