| Valeriana officinalis L.belongs to the family of Valerianace,It is a herbal traditional Chinese medicine which was long used to treat illness such as disquiet,insomnia and aches of various kind.Modern research shows that,it has a great potential capability of curing cardiac arrhythmia.In order to find out how it works,we have studied the anti-arrhythmia effective substances,including pharmacological selection of the effective extracts,constituents isolation from the effective extracts and GC fingerprint.Pharmacological method was adopted to compare effect of different extracts.Result as the water-extract and oil have obvious anti-arrhythmia effect induced by trichloromethane,the lower dose of ethyl acetate extract has similar effect,and the n-BuOH extract was correspondingly less effective.The water-extract and the n-BuOH extract have obvious anti-arrhythmia effect induced by aconitine,while the other extracts can be effective at certain does.GC-MS method was adopted to have identified 51 constituents from the oil extract;The methods of solvent and chromatography were used to get 10 monomeric compounds from the EtOAc fraction and n-BuOH fraction,and all of them were elucidated by spectroscopic spectrum as prinsepiol-4-0-β-D-glucoside(â… ),8-hydroxypinoresinol-4-o-β-D-glucoside(â…¡), rulepidol(â…¢),β-sitosterol(â…£),valerenicacid(â…¤),kanokosideA(â…¥), Orivalerianol(â…¦),Behenic acid(â…§),Monovalerianester A(â…¨), pinoresinol(â…©);Compondâ…¢,â…¦,â…§andâ…¨were discoveredfirstlyfrom this plant,and compondâ…¦,â…¨are 2 new natural products.Fingerprint is a good technology to evaluate the quality of herbal medicine.The main active fraction was extracted by SFE,and the GC Fingerprint was studied,27 peaks were assigned as common peaks in the spectrum.The analysis method has good credibility and stability.The research provided certain scientific parameter to control the quality of Valerian. |
