Objective To investigate the effect of Erigeron breviscapine on the apoptosis and the express of bax,bcl-2mRNA in kidney of diabetic rats.Methods Normal male rats were selected.Rats were randomly divided into three groups:normal group(N),diabetic group(DM),breviscapine treatment group(EB).DM and EB group were induced by injecting streptozotocin(STZ)to become the model of the diabetic. Erigeron breviscapine was injected into rats in EB group by the coeliac cavity(20mg.kg-1d-1).Rats of three groups were killed at 2weeks and 6weeks after giving medicine.Apoptosis were identified by terminal deoxy nucleotidyl transferase mediated d-UTP nick end labeling(TUNEL).Expression of bax and bcl-2mRNA in kidney were detected by real-time quantitative with SYBR GreenⅠ.Results①Compare with normal group,the apoptos cell in tubules of diabetic group and breviscapine group is increase obviously at 2and 6 weeks,diabetic group is more than breviscapine group.Apoptosis in glomeruli is not detected at 2 weeks,is detected rarelly at 6 weeks.②Compare with normal group,the expression of bax,bcl-2mRNA of diabetic group is in increase obviously,the ratio of bax/bcl-2 is in increase. Compare with diabetic group,the expression of bcl-2mRNA of breviscapine group is in increase,baxmRNA is in decrease,the ratio of bax/bcl-2 is in decrease. Conclusion Breviscapine has renal protection from diabetic nephropathy,partly through inhibiting apoptosis by regulating bcl-2 and bax expression. Objective To investigate the effect of Erigeron breviscapine on the renal function, structure and the expression of transforming growth factor-batel(TGF-B1)in diabetic rats.Methods 70 normal male rats were selected.Rats were randomly divided into three groups:normal group(N),diabetic group(DM),and br-eviscapine treatment group(EB).DM and EB group were induced by injecting streptozotocin(STZ)to become the model of the diabetic nephropathy.Erigeron breviscapine was injected into rats in EB group by the coeliac cavity.Normal Saline was injected into rats in N and DM groups by the same cavity.Rats of three groups were killed at 2weeks and 6weeks after giving medicine,then blood specimen was collected to examine blood glucose(Glu),serum creatinine(Cre),blood urea nitrogen(Bun)and TGF-B1 concentrations,The changes of the glomerulus area(AC)and volume(VC)in each group were observed.Rat renal tissue were also collected to examine the glomerular membrano-proliferation and the renal interstitial arteriolosclerosis.Results①Comparison of TGF-B1:diabetic group>breviscapine treatment group>normal group.②Comparing with normal group,the weight of diabetic group and breviscapine group decreased,renal index,AC and VC increased.Comparing with diabetic group, the weight of breviscapine group was increased,renal index,AC and volume were decreased(P<0.05).③Comparison of Cre and Bun:diabetic group>breviscapine treatment group>normal group.Compare with diabetic group,the level of Cre and Bun were lower.Cre decrease was more obvious in 2 weeks(P<0.01).④The glomerular membrano-proliferation and the renal interstitial arteriolosclerosis are diabetic group>breviscapine treatment group>normal group.Conclusion Erigeron breviscapus can repress the expression of serum TGF-β1,repress kidney overgrowth and delay renal function to worsen in diabetic rats.
