Objective: To investigate the effect of gemcitabine on the growth inhibition and induction of apoptosis in osteosarcoma cell line: MG-63.Methods: The morphological changes were examined by phase contrast microscope ;cell growth rates were assessed by MTT bromide colorimetric assay. The apoptosis of MG-63 was measured by flow cytometry(FCM) with Annexin-V-FITC assay and DNA Ladder.Results: Gemcitabine could significantly inhibit cell growth and the inhibitory effect was in dose-and time-dependent manner evidenced by MTT assay and morphological changes. The apoptotic index measured by FCM increased correspondingly with the concentration of gemcitabine and the reaction time. Treating MG-63 with different concentrations (0, 0.1,1.0,10.0μg/ml) of gemcitabine for 48h, the apoptotic index was 5.56%,59.82%,52.90%,60.20%, respectively; the death index was 3.15%,8.46%,18.15%,17.43%, respectively. Besides DNA Ladder could be demonstrated on DNA electrophoresis.Conclusion: The apoptosis of osteosarcoma cells, MG-63 cell line, was successfully induced by gemcitabine, and that was dose- and time-dependent.
