Amphibious Two Elixir Experimental Study In Rats With Adriamycin Nephrosis

Objective:To study theraputic effects and mechanism of ShuiLuerxiandan on Adrianmycininduce nephrotic syndrome(NS)in rats.Method:Fifty male Wistar rats were selected and randomly divided into five groups.Tenrats were used nomal group,and the the remaining rats were given intranvenouseinjection of Adrianmycin (6.0mg/kg) to establish modle of NS.The modle of NS ratswere divded into four groups:the molde group (n=10), high shuiluerxiandan group(n=10)treated with high shuiluerxiandan, low shuiluerxiandan group(n=10) treatedwith low shuiluerxiandan and bennazepril group(n=10) treated with bennazepril for21 days respectively.Normal group and the model group were given 0.9% sodiumchloride.Three weeks after treatment, the general condition of rats,weight,24h urineprotein quantity, urine albumin, urineβ₂-microglobulin, urine immunoglobulin,serumalbumin, serum total protein, serum crearinine, blood urea nitrogen,cholesterol,triglyceride, tumor necrosis factor—α, hyaluronic acid and patholoficalchanges of renal tissues were observed.Results:24h urine protein quantity increased significantly 7 days after the injection ofAdrianmycin.After 21 days treatment,the groups treated showed decreasingsignificantly in 24h urine protein quantity, urine albumin, urineβ₂-microglobulin,urine immunoglobulin,cholesterol, triglyceride, tumor necrosis factor—α, hyaluronicacid,and increasing in serum albumin, serum total protein(P＜0.01,versus the modelgroup).High and low shuiluerxiandan groups got better theraputic effects ofdecreasing in cholesterol, triglyceride and increasing in serum albumin, serum totalprotein (P＜0.05,P＜0.01,compared with the bennazepril group). TNF—α,HA in thegroups treated with Shuiluerxiandan decreased significantly (P＜0.01,versus themodel group).Pathological studies showed obvious fusion of footprotrusion withoutmarked thickening of glomercular basement membrance in the modle group.Thefusion of partial footprotrusion was observed in the groups treated. Conclusion:The results showed:the nephrotic syndrome models were induced byintravenouse injection of Adrianmycin,all treated groups could get the theraputiceffects,the theraputic effects of decreasing cholesterol, triglyceride and increasing inserum albumin, serum total protein in the Shuiluerxiandan were better than thebennazepril group, TNF—α, HA in the group treated with Shuiluerxiandan decreasedsignificantly compared with the model group,which suggested the treatment withShuiluerxiandan could get better theraputic effects on nephrotic syndrome byeffecting the cell-mediated immunity mechanism.
