Preliminary Study. Fuzheng Blood Circulation Ecm Metabolism In Renal Interstitial Fibrosis In Rat Renal Tubular Interstitial

Objective: The renal tubulointerstitial fibrosis rat animal models were formed by Unilateral Ureteral Obstruction. To observe the expression of the fibronectin (FN) in kidney in the process of renal tubular damage and interstitial fibrosis in UUO rats. To investigate the therapeutic effects and its mechanisms of Chinese herbs with supplementing qi, activating blood circulation and eliminating blood stasis therapy in the process of renal tubular damage and interstitial fibrosis in UUO rats.Methods: Thirty male Wistar rats were randomly divided into three groups: the control group (Sham group), the model group (UUO group), Chinese herbs treatment group (A group). The 2ml distilled water or the same amount, of Chinese herbs were administered by gavage. beginning three days before Unilateral ureteral Obstruction preparation and continued through twenty-one days after Unilateral Ureteral Obstruction operation. Sera and the kidney tissues were collected from each group on the twenty-first day. BUN and Scr were. measured. The rats' left kidneys were fixed with Formalin, and the slices were disposed in routine method, dyed with Hematoxylin-Eosin or Masson staining, and observed with computer image analysator.It was carried out to measure the level of tubulointerstitial damage by Masson staining. The expression of fibronectin (FN) in kidneys was assessed by immunohisto- chemistry.Results: 1. Renal function: The level of BUN and Scr in UUO group were significantly highter than in Sham group. There was significant difference betweem Sham group and UUO group (P<0.05, P<0.05). Compared with UUO group, the level of BUN and Scr in the Chinese herbs treatment group were significantly lower (P<0.05, P<0.05). 2. Pathology: The renal tissue were stained with HE. The renal structure of the Sham group showed no pathological changes. But in UUO group, it was shown that there was tubular epithelial cells denaturalization, renal tubular expand or atrophy, interstitial macrophage and monocyt infiltration, interstitial expand, and there was no glomeruli change. While the change in the Chinese herbs treatment group was slighter than that in UUO group.The renal tissue Masson stainning showed that there was interstitial expand, fibrous tissues proliferation and there was no glomeruli change in UUO group. While the change in the Chinese herbs treatment group was slighter than that in UUO group.Pathologic irfiage analysis shows that the renal interstitial fibrotic area expanded markedly in UUO group. There was significant difference betweem Sham group and UUO group (P<0.05), Compared with UUO group, fibrotic area of the Chinese herbs treatment group was decreased markedly (P<0.05).3.Immunohistochemistry: The expression and distribution of FN: There was a weak expression in Sham group, while in other group the expression of FN increased in renal interstitial. Image analysis shows that the positive area and density of FN were highter significantly than that in Sham group. There was significant difference betweem Sham group and UUO group (P<0.05, P<0.05). Compared with UUO group, the positive area and density of FN in the Chinese herbs treatment group decreased markedly. There. was significant difference betweem the Chinese herbs treatment groupgroup and UUO group (P<0.05, P<0.05)4.Correlation analysis: Correlation of CTGF and fibrotic area was positive correlation. Correlation of fibrotic area and BUN was positive correlation, Correlation of fibrotic area and Scr was positive correlation.Conclusion: 1. The expression of FN upregulated significantly in kidney in the process of renal tubular damage and interstitial fibrosis in UUO rats. 2. Chinese herbs with supplementing qi, activating blood circulation and eliminating blood stasis therapy can downregulate the level of BUN and Scr in Unilateral Ureteral Obstruction model rats, and protect renal function.3. Chinese herbs with supplementing qi, activating blood circulation and eliminating blood stasis therapy can suppress the upregulation of FN expression in kidneys of Unilateral Ureteral Obstruction animal model rats and decrease their interstitial fibrotic area. It is inferred that it may be one of mechanisms of Chinese herbs with supplementing qi, activating blood circulation and eliminating blood stasis therapy suppressing renal tubular damage and interstitial fibrosis.
