Objective: To observe the effect of alendronate and raloxifene on the lever of biochemical indices, histomorphometry and bone density in the experimental osteoporosis and periodontitis rats models. To evaluate the anti-osteoporosis effect of alendronate and raloxifene on bone in rat model.Methods: Fourty-eight female SD rats aged 3 months were divided randomly into six groups. The osteoporosis models were established by ovariectomy and the periodontitis models were established by dental ligature around the dental cervix of maxillary upper first molar, with 2mm diametric orthodontia ligature wire. The experimental groups were given medicine by oral after fifth day of ovariectomy and dental ligature, each day for 3 months. The blood was then sampled from the arterial ophthalmic under ether anesthesia. The blood was allowed to clot to separate the serum immediately for determination of calcium(Ca),phosphate(P),alkaline phosphatase(ALP).Then the rats were sacrificed under extra ether anesthesia. And the left femora and dental-periodontal tissues were removed, they were fixed in 10% Formaldehyde for 24h for determination of bone density and histomorphometry. All statistics are represented in the form of -x±s, SPSS11.5 statistic software was used to tackle the datum, t inspection and variance analysis are used to analyze.Results: 1.Body weight changes:in all the 6 groups body weight increased from week 1 to week 12.In the ovariectomy group,the body weight increased greater in comparison with that in the raloxifene group and Sham group (P<0.05). 2.Effects on calcium: in the raloxifene group and ovariectomy group(ovariectomy group and ovariectomy plus dental ligature group),the content of calcium was significantly greater in comparison with that in the Sham group (P<0.01) ;in the ovariectomy plus dental ligature group,the content of calcium was significantly greater in comparison with that in the alendronate group(P<0.01).And in the ovariectomy plus dental ligature group,the content of calcium is significantly greater in comparison with that in the raloxifene group(P>0.05).As compared with the raloxifene groups, the values of alendronate groups significantly reduced biochemical indices. 3.Effects on phosphate:In all the 6 groups the content of phosphate had no significant different(P>0.05). 4.Effects on alkaline phosphatase:in the ovariectomy group and dental ligature group,the content of alkaline phosphatase was significantly greater in comparison with that in the Sham group(P<0.01);in the ovariectomy plus dental ligature group,the content of alkaline phosphatase was significantly greater in comparison with that in the alendronate group and raloxifene group.( alendronate group ,P<0.01;raloxifene group,P<0.05)。As compared with the raloxifene group, the values of alendronate group were significantly reduce biochemical indices. 5.Effects on the BMD of the femurs and maxillary:in the ovariectomy group the BMD of the femurs and maxillary were lower than Sham group(P<0.05); the BMD of the femurs and maxillary in alendronate group significantly taller than ovariectomy plus dental ligature group(P<0.01).As compared with the raloxifene group, the values of alendronate group were significantly improved on bone density. 6.In histomorphometry, there were little inflammation in gingivitis and alveolar crests resorption of the medicine groups.Conclusions: 1 There is maxillary and system osteoporosis in the ovariectomy group.2 The periodontitis and alvelor absorption in osteoporosis groups are obviously sever than normal groups.3 Both ALN and RLX can prevent osteoporosis,however, ALN can do better in preventing bone loss to reduce osteoporosis in ovariectomized rats and prohibitingl alveolar bone resorption.
