In order to study the influence of garoderma lucidum polysaccharides to NF-κB which expressed in epilepsy model cell. Methord: 30 neonatal Wistar mice were killed to get hippocampal neurons which were cultured for seven day. hippocampal neurons were divided into 3 groups at random: Garoderma lucidum polysaccharides,epilepsy model group and blank group. immunofluorescence reaction was took in different time:0.5 hour,1 hour,6 hours,12 hours. After closing by serum NF-kB/p65 antibody was added. Then it was washed by phosphoric acid buffer liquid before adding FITC-IgG antibody to hatch. Finally it was dyed by propidium and observed with the fluorescence microscope. NF-κB/p65 subunit are marked green by FITC-IgG antibody. cell nucleus are marked red by propidium. applying the immunity organization chemistry to examine each variety of related index sign. Results: culturing hippocampal neurons of neonatal mice are successfully. they grow at the seventh day to prepare modle of cell epilepsy. It is proved that the expression of NF-κB in nuclear are more than garoderma lucidum polysaccharides and blank contrast group. It hint that hippocampal neurons have been active. the expression of NF-κB in garoderma lucidum polysaccharides are less than in epilepsy model group. Conclusion: According to the results, the method of cultured hippocampal neurons which use culture medium and medicine is available, hippocampal neurons grow well,.pure hippocampal neurons add up to 90%.providing better model study experiment of correlative hippocampal neurons . it is testified that active NF-κB in nuclear is relate to epilepsy pathogenesis. The expression of NF-κB in nuclear is sign of active hippocampal neurons. garoderma lucidum polysaccharides can effectively reduce cell [Ca2+] overload of the neuron cell effectively, indirectly inhibit the expression of NF-kB in nuclear to reduce excitability of neurons.
