| Objective:Chronic renal failure (CRF) is damage to kidney which is caused by various kinds of idiopathic or successive renal disease. Half-backed renal function leads to a series of clinical symptoms which are caused by decompensate. The renal pathological changes develops slowly but it is one-way and irrecoverable. It evolves as terminal renal disease (Uraemia) at last.The pathogenesis of CRF can be unpuzzled with the theory of Chinese medical that it is correlated with asthenia, turbid, and stagnancy. Yi Shen Granule can enrich qi , nourish yin, remove blood stasis and clear away turbidness. Yi Shen Granule has a definite effect on CRF which is caused by asthenia of qi of the spleen and kidney, damp-turbid, damp-heat or blood-stagnancy. We studied on the curative effect of Yi Shen Granule with means of modern pharmacology , tried to explore the curative mechanism of Yi Shen Granule.Methods:Rat models of chronic renal failure were made by cut 5/6 kindney or fed with feedingstuff which contains adenine(0.75%) for 4 weeks. Experimental treatment was applied by giving Yi Shen Granule and the curative effect of Yi Shen Granule on ongoing pathological changes of CRF was studied. According to the data of CRF rats in weight, water wastage, hemanalysis(RBC,HGB,HCT,MCV,MCH), urinalysis(volume, specific gravity, pH, proteinuria), blood serum analysis(BUN,Cre,ALB,TP), kidney weight coefficient and kidney morphology ( the grade of expansile renal tubule and the diameter of glomerulus ),the pharmacological effect of Yi Shen Granule could be ascertained. We used excretion test of phenolsulfonphthalein to compute renal plasma flow (RPF) and measured the blood viscosity of CRF rats to validate the function of removeing blood stasis of Yi Shen Granule. Swimming test and cold-resistant test for mice were made to study the effect on antiirritability. The eliminated rate of carbon granule and the content of hemolysin in serum in low-immune mouse were measured to explore the effect of Yi Shen Granule on immune ability. Results:For CRF rats which made by fed with feedingstuff contains 0.75% of adenine for 4 weeks, the weight of model control group rats were lighter than that of Yi Shen Granule groups rats. The water wastage of Yi Shen Granule groups rats were less than that of model control group rats. Compared urine volume, the ejectable quantity of Na, K, Cl and proteinuria in 12 hours , the data of Yi Shen Granule groups rats were less than that of model control rats. The specific gravity of Yi Shen Granule groups rats was higher than that of model control rats. The numerical value of RBC, HGB, HCT, MCV, MCH in model control group was lower than that in Yi Shen Granule groups. The content of BUN and Cre in serum of model control groups rats were higher than that of Yi Shen Granule groups. The results of excretion test of phenolsulfonphthalein showed that the RPF of Yi Shen Granule groups rats was larger than that of model control group rats. After administerded for 30 days, all rats were killed and anatomized. The nephric exterior of model control group rats was offwhite, instead, that of Yi Shen Granule groups rats showed red grain in offwhite exterior. There were tumefaction of tissue and sediment of adenine in all kidney. Pathologic results showed that there was dilation of renal tubule in different grades, the grade of dilation of renal tubule in high dose Yi Shen Granule groups rats was lower.Administerded Yi Shen Granule to CRF rats which made by cut 5/6 kindney for 42 days, the content of ALB, TP, BUN and Cre in serum were measured after 28 days. Compared with model control group, the results indicated that high dose and mid dose Yi Shen Granule could inCrese the number of ALB and TP and depress the concentration of BUN and Cre in serum. At same time, the concentration of BUN and Cre in model control group rats was higher than that before 28 days. At No.42 day, all rats were killed, anatomized and measured the viscosities of whole blood and plasm. Compared with model control group, the results indicated that high dose and mid dose Yi Shen Granule could lighten compensatory hypertrophy of glomerulus, the viscosities of whole blood and plasm in Yi Shen Granule groups rats were low.For normal mouse, Yi Shen Granule could prolong the swimming time of mouse, the survival in Yi Shen Granule groups was more than that in model control group in same cold condition. For low-immune mouse, The eliminated rate of carbon granule in Yi Shen Granule groups rats was faster than that in model control group rats, it showed that phagocytosis was boosted up, the absorbency of upside liquid in Yi Shen Granule groups rats was more than that in model control group rats in hemolysin mensurated test, it indicated that more antibody was come into being by giving Yi Shen Granule. Conclusion:Yi Shen Granule could protect renal functions, depress BUN and Cre, ameliorate a series of clinical symptoms which were caused by decompensate, restrain hypertrophy of the remains kidney, decrease the incidence of glomerular, delay the development of CRF, strengthen anti-stressor and immune ability. The results attested that Yi Shen Granule has the functions of enriching qi , nourishing yin, removeing blood stasis and clearing away turbidness. |
