Objective:To investigate the localization of AQP9 and its mRNA in different position of the rat digestive tract, including esophagus, forestomach,glandular stomach, proximal small intestine, distal small intestine,proximal colon,distal colon and liver,so as to elucidate their function in different sites of the digestive tract.Methods:10 healthy SD rats(BW=180~220g)were perfused with 0.9% saline and 4% PFA through the ascending aorta. Then esophagus, forestomach, glandular stomach, proximal small intestine distal small intestine, proximal colon, distal colon and liver were removed and sliced. The IHC and ISH methods were used to study AQP9 and AQP9mRNA expression. After that, the imaging analysis of their expression was made by means of Computer-aided Image Analyzer.Results:AQP9 was expressed in the rat hepatocyte,columnar epithelium of small intestine and colon, intestinal gland, goblet cell and parietal cell.There were few positive signal in the mucosa of esophagus and forestomach.
