Object To study the mechanism of TGF-β1 in postmenopausal osteoporosis. To illuminate the herbs of invigorate the kidney to regulate it,providing experiment base for prevention and cure the disease in clinical. Methods Common Wister rats were made postmenopausal osteoporosis after unsex way,which divided normal group, model blank group , herb complexed group , gushukang group, nilestriol group, were lavaged saline of same volume in 12 weeks. Survival rats were executed in 24h after the last lavaged. Samples were determined and analysed. E2; LH; FSH;BGP were determined by radioimmunosaay (RIA) ;ALP was detected by IFCC;Uterus were weight with torsion balance after 4% PFA perfusion and counted exponent. At the same time, left legs were determined by bone density machine. TGF-β1 was determined with immunohistochemical SABC method in Kidney. Results 1)model group has osteoporotic character, estrogen decrease, uterus contract greatly, bone density dreased. Bone metabolism has the character of bone metabolism and bone absorption increasing. 2)Herb can increase femur bone density, compared model group (P<0. 05). 3) nilesteiol can increase uterus exponent. Herb and gushukang group can increase uterus exponent, but hasn' t significant difference (P>0.05) .4) Immunohistochemical display TGF-β1 expression increase in model group, much brown masc product in tubule, and distributed like trabs. statistical analysis indiscated, herb complexed decreased TGF-β1 expression ; TGF-β1 expression , E2 and bone density were negative correlation . Conclusion blood serum 1)E2 decrease greatly in postmenopausal osteoporosis.2) TGF- β1 signal transduction molecule expression increase in postmenopausal osteoporosis. 3) The herbs of invigorate the kidney can prevention and cure postmenopausal osteoporosis by regulating TGF-β1 signal transduction.
