Malignant tumour is a common disease, which threatens human life severitily. It is called the first killer of human health. Although the cure of malignancy has gained quite great progress during the recent years, the clinic using drug has still remained in the published data and clinic experience. Different tumour sufferers were treated with fixed chemotherapy method, which inevitably brought blindness of using drug and endurance of the drug. Therefore, the treated method would induce low curative or useless effect. So the choice of chemotherapy method is the key of success or failure in treatment. As a result, it is a hotspot to search for a good experimental project of the tumour drug sensitivity nowadays. The filtration and appraise of the drug depends on the reasonable methods and system. The purpose of the experiment is to build a drug appraise model in the cellular and animal level in order to appraise the effect of several antitumor drug.In the cellular level, the cytotoxic effect of antitumor drug (Unite gene p53/p21, taxol or Taxol derivative, adriamycin) on human laryngeal cancer Hep-2 cell line was examined by MTT assay, morphological observation, gel electrophoresis of DNA, and cell morphology. The results showed that antineoplastic drug inhibited the proliferation of Hep-2 cells in a time and dose dependent manner in vitro. Chromation condensation, nucleic fragmentation and apoptic body formation were found in the cell morphologyThe three drugs could inhibit the proliferation of Hep-2 cells in a time and dose dependent manner in vitro. The cytotoxic effect of antitumor drug on Hep-2 cell was achieved by inducing apoptosis.In the animal level, the inhibited effect on the mouse sarcoma S-180 of the three antineoplastic drugs was evaluated, using the tumour bulk and suppressive tumour ratio as the appraise guideline. The investigation results showed the obvious inhibited effect of the three drugs.
