| Probiotics play an important role in health improving and disease preventing. The purpose of this research firstly is to screen the probiotic bacteria from a great deal of lactic acid bacteria, and identify them so as to lay a foundation for studying and developing probiotic products with independent knowledge property right.This study is divided into three steps: first it focuses on the function characteristics of lactic acid bacteria so as to obtain good probiotic bacteria, and identifies the probiotic bacteria, at last the process characteristic of the probiotic bacteria.The ability of the acid- and bile-tolerant is studied with the lactic acid bacteria isolated. All of the 15 strains can exist at pH3, 0.3% bile. Only 10 strains can grow for 5 hours at pH2, 0.3% bile, and Q grows best, with the best ability of the acid- and bile-tolerant. Only Q can grow at pH3, 0.2% bile.The ability of inhibition from Escherichia coli and Helicobacter pylori is studied with the lactic acid bacteria isolated. All strains can inhibit from Escherichia coli ,and Q has the strongest effect of inhibition from Escherichia coli, with inhibit circle 12.6mm exceeding the type strain BB-12's 8mm.Most strains can inhibit from Helicobacter pylori,B has the strongest inhibition effect with the inhibit circle 13mm, and Q is at the second place with the inhibit circle 12.7mm, comparing with BB-12's 9.8mm.The ability to adhere to human intestinal cells in vitro is studied with the lactic acid bacteria isolated. Strain A shows the best effect of adherence to human intestinal cell Caco-2 with the average adherence amount of 18.7±5.92 per cell, followed by Q's average adherence amount of 12.9±2.74 comparing with BB-12's 10.7±2.46. When adhering to Caco-2 for 2 hours with Lactobacillus then with Escherichia coli, Escherichia coli can be inhibited thoroughly by strain Q and E. When adhering to Caco-2 cell with Escherichia coli and then with Lactobacillus for 2 hours, the average adherence amount is almost the same as that of contrasted Escherichia coli.The ability of anti-oxidation is studied with the lactic acid bacteria isolated. It is found that when the mice ate the fermentation product with strain Q for 8 days, their ability of SOD was enhanced and the content of MDA reduced. As a result, strain Q performs the better ability of anti-oxidation. |
