Objective: The study intended to explore the main environmental risk factors of esophageal carcinoma ,and the relationship of the genetic polymorphisms of NQO1,MGMT and susceptibility to esophageal carcinoma(EC),and the gene-gene or gene-environmental interaction ,so as to provide theoretical evidences for making the general preventive measures andrevealing etiological mechanisms for esophageal carcinoma in Changzhi Shan'xi province.Methods: A 1:1 matching case-control study which included 201 cases of esophageal carcinoma and 201 cases of controls, comed from Heping Hospital and Heji Hospital of Changzhi from Aug 2001 to Jul 2005. Cases were definited esophageal carcinoma by gastroscope and pathology means; Controls had no tumor history and relative disease of esophagus in the same period. All cases were investigated by a questionnaire about the related factors of digest systemic disease and drawed elbow'venous blood 3 ml to detect genotype, using polymerase chain-restriction on fragment length polymorphism (PCR-RFLP). Statistics analysis adopt SPSS11.5 statistical packages, All data were analyzed byconditional logistic multiple factor regression analysis andχ2-test et al.Result: 1.Analysis of multiple variables logistic regression showed that smoking,liking to eat hot,hard diet,pickled food, fierce mind stimulation and family history of esophageal carcinoma were main risks to EC in Changzhi area, OR(95%CI) were1.19(1.16~1.32),2.19(1.24~3.86),2.14(1.25~3.68),2.01(1.15~3.54),1.74(1.01~3.00),2.69(1.23~5.90)respectively.Intaking more animal protein was the protective factor to EC, OR(95%CI) was 0.57(0.40~0.83).2.There wasn't significant association between the genotype distribution of NQO1,MGMT and age,sex,smoking,eating pickled food;NQO1 have three kinds of genotype : mutation homozygote,mutation heterozygote and wild homozygote,the requencies of them were 20.90%,65.17%,13.93% in case group, 16.42%,54.23%,29.35% in control group respectively,there was significant difference between two groups(χ2=14.14,P =0.00).The genotype frequencies of MGMT , were 47.76%,28.86%,23.38% in case group and 43.28%,42.29%,14.43% in control group respectively,there was significant difference between two groups(χ2=9.80,P =0.01).3. The odds risk of NQO1 mutation homozygote and mutation heterozygote to wild homozygote were significant increased, OR(95%CI) were 2.53(1.51~4.25),2.68(1.41~5.09) respectively.And the risks of MGMT mutation genotypend and mutation heterozygote were signification increased to wild genotype, OR(95%CI) were 2.53(1.51~4.25),和2.68(1.41~5.09) respectively. 4.
