| ObjectivePeripheral nervous trauma (PNT) is a common severe disease of orthopedics department, which cause the body serious crash, stretch, twist,tear in traffic accident, high fall and heavy crash or injured by high-speed bullet, fracture, dislocation, produced injury, narcosis, cranimation and sopor, cause peripheral nerve different injury. INT can cause hematomyelia, dropsy, necrosis, capsular space, gummosis and so on, which can lead to neuron deprivation, demyelination of neurite. Clinical symptoms mainly manifest function disorder of movement, sense, reflection, and musculus sphincter below injury limb. Schwann cell, locate in Peripheral nervous system, can change microenvironment post-PNT, prevent secondary lesion as well as form guidance passage of regenerating axonal. Schwann cell can also secrete massive neurotrophic factor and cytokine which might promote functional recovery of limbs. YaoTongDing Injection (YTDI) is composed by Chinese herbs which can promote qi to active blood. In this experiment, YTDI was made into containing drugs serum which was used to cultivate Schwann cell. Cell proliferation was observed in this experiment. All these data can provide evidence for investing the mechanism of treating spinal injury with Chinese herbs and help researchers choose proper dose for YTDI.MethodsIn cell identify experiment, the sciatic nerve from SD rats was digested by pancreas and collagenase and then inoculated to culture flask, the identification of Schwann cell subcultured was showed by indirectly immunocytochemistry with S—100 protein McAb, and compare with the fibroblast by the same method.In PH value determination experiment, the PH instrument show YTDI's PH of different concentrations and compare with 10% calf serum DMEM cell culture medium.In cell multiplication experiment, the subcultured Schwann cell was diluted and digested, then inoculated to culture board , randomly divided into experimental groups and comparison group. The YTDI of different concentrations were affiliated into each experimental group respectively, and then observe the cell proliferation change of each group now and after 3, 5, 7, 9, 11 days in vitro, and experiment result was record after MTT analysis.Numerical data are expressed as the mean ± standard deviation.
