| This paper reports the primary anticancer activity of Injectable AIDI (gelsiccation) in vitro and in vivo. The preparation safety for the injection of Chinese materia medica and toxic action are also investigated.The studies demonstrate that AIDI inhibited the proliferation of human hepatoma BEL7402 ,human cervical carcinoma HeLa ,human melanoma A375-S2, human gastrv-carcinoma SGC-7901 and mice fibroma sarcomatosum L929 in a dose- and time-dependent manner by MTT colorimetric assay.In vivo, the tumor growth of mice with H22 was inhibited compared with the control group after animals were treated with AIDI i.v. at 0.972 g·kg-1 , 1.943 g·kg-1, 3.886 g·kg-1 for 7 days, and the average tumor inhibition rates of AIDI at the three doses were respectively 28.39%, 41.02%, 48.37%. The tumor growth of mice with S180 was inhibited also, and the average tumor inhibition rates of it at the three doses was 32.02%, 44.20% and 49.74%, respectively. After C57BL/6 mice were treated with AIDI iv. at 0.972 g·kg-1, 1.943 g·kg-1, 3.886 g·kg-1 for 12 days, the average Lewis lung tumor inhibition rates of AIDI at the three doses were respectively 20.04% , 40.44% and 50.75%. In addition, AIDI showed significant synergism effect with CTX in the growth inhibition of S180 in mice.The possible mechanism of antitumor effects of AIDI maybe associate with inducing apoptosis.AIDI in coordination with CTX increase the activity of NK cell and immune function of T lymphocyte subgroup in L7212 tumor -bearing mice. AIDI stimulate the growth of bone marrow cell in normal mice, and even rise the numbers of white blood cell after the treatment at 1.943 g·kg-1 in H22-bearing mice. In the study of carbon particle clearance in S180 tumor-bearing mice, it was demonstrated that Phagocytosis index (k) was significant risen at 1.943 g·kg-1 dose compared with CTX group. These results indicated that AIDI increase anti-tumor immunity.The result of safety experiment shows that AIDI hasn't hemolysis reaction to red blood cell from rabbit, and its muscular tissue stimulus and blood vessel stimulus in rabbit were slight and reversible. The test of initiative hypersensitive reaction suggested that AIDI had no allergization in cavia cobaya. and PCA test showed no IgE antigenic effect in rats.The chronic toxic test showed that toxic reaction in rats is reversible and no relay toxic... |
