| Objectives: To evaluate the antibacterial activity of Mezlocillin / tazobactam (MLN/TAZ) (1:0,1:1,2:1,4:1,8:1)in vitro and MLN/TAZ (1:0,2:1,4:1) in vivo.Methods: Minimal inhibitor concentrations (MIC) of MLN/TAZ (1:0,1:1,2:1,4:1,8:1) were determined by agar double dilution of 421 strains of clinical organisms, and compared with Cefotaxilne / tazobactam (5:1), Cefoperazone / tazobactam (4:1)and Ceftazidime, minimal bactericidal concentrations(MBC) were measured by broth dilution method, and bactericidal curves were produced by counting of mattress drop method . The effects on MIC of changing the pH values,concentration of serum protein and the quantity of inoculated bacteria were observed.The models of acute septicemias with E.Coli 1252 producing ESBLs were established in mice, and the optimum ratio of MLN/TAZ was explored by this model.Results: The antibacterial activity of MLN/TAZ (1:1,2:1,4:1,8:1) against clinical isolates was better than MLN alone and CAZ ,and similar... |
