Aconitum Aconitum Boiling Time Dose Toxicity Studies

Objective: Revealing the relativity of different dectocting time (DT), administration dosage (AD) and the toxicity of Radix Aconiti (RA) and Radix Aconiti Kusnezoffii(RAK).Methods: Employing Uniform Designed technique, we dectocted RA and RAK at identical conditions with different DT of 15 minutes, 30minutes, 1hour, 2hours, 3hours, 4hours and 6hours, according with 6times, 12times, 24times, 48times, 72times, 96times, 120times of clinic AD .We randomly set the male SPF rats into 9 groups . All rats except normal contrast group were made BiZheng animal model. On the 30th day, we took the femoral vein blood of 2/3 each groups animals, and did a series of hematology and blood biochemistry test, anatomied, histopathology test. Then we observed the rest animals in 15 days. Test methods and indexes are same with the first time. The outcomes of Regression Analysis revealed the relativity between DT, AD and the toxicity of RA, RAK.Results: 1. Outcome of taking RA 30-day: (l)The feet tumefaction: Compared with model group, taking RA groups' feet tumefaction was alleviated in varying degrees. The best DT and AD are 30 minutes and 120 times for anti-inflammatory. (2)Pain threshold value: Taking RA groups rats pain threshold value was improved progressively, and it approached the normal level at the 30th day. The best DT and AD are 30 minutes and 120 times for analgesia. (3)Weight: The difference between each groups was not statistics value. (4)Hematology test: All outcome of hematology all rats were normal. The difference between each groups was not significant in biology. (5)Blood biochemistry test: Compared with normal group, Crea value of taking RA groups except 2-hour group and Urea value of 6-hour group risen obviously. The DT and AD of maximal toxicity are 30 minutes and 120 times. (6)0rgan coefficient: Compared with normal group, the kidney organ coefficient of 1-hour group risen obviously. The DT and AD of maximal toxicity are 30 minutes and 120 times. (7)Histopathology : liver and kidney of few rats has slight metastasis in 15-minites group and 30-minites group. RA reversibility observation: The feet tumefaction and pain threshold value of each groups approached the normal level at the 15th day.
