Purpose: To study the estrogenic properties of the Extracts from Cajanus cajan L (ECC) in osteoblastic activity and bone resorption, we identify its effects in HOS TE85 osteoblast-like cells, marrow-derived osteoclast-like cells and ovariectomy (OVX)-induced bone loss rats. Methods:1) In vivo:2 weeks after the ovariectomy, drugs No. 32—1 were given to the rats for 8 weeks through stomach. Rats were sacrificed and bodies and uterus were weighted, serum estradiol (E2), FSH, LH concentrations were measured by radioimmunoassay. Femoral morphology was observed by HE staining.2) In vitro:Ⅰ After the treatment of ECC (10-7g/ml, 10-8g/ml, 10-9g/ml) for 48 hours, by using MTT assay to test ECC's effect on cell proliferation, 3H-proline incorporation to investigate the formation of collagen, and by measuring alkaline phosphatase (ALP) activity to evaluate bone formation on HOS TE85 cell.Ⅱ Test the ECC (10-8g/ml) 's long term (18 days) effects on promoting mineralized bone-like tissue formation in HOS TE 85 osteoblast like cells by Alizarin red S staining.Ⅲ Bone marrow cells were cultured to examine ECC effects on the derivation of osteoclast cells(OLCs), which were stained by Tartrate-resistant acid phosphatase (TRAP).Results:1) In vivo:By HE staining, we found improved femoral morphology of OVX rats after thetreatment of ECC. 60% of the bone loss cases were ameliorated after the treatmentof high dose of ECC (200 mg/kg · d), 30% of the bone loss cases were ameliorated...
