Chinese forest frogs (Rana Chensinensis), called toad in usual, are a kind of amphibian. Chinese forest frogs only live in China Northeast and have high economical value. At the present, the researches on Chinese forest frogs mostly focus on the oviducts of female frogs, little report are about the functions of other parts in Chinese forest frogs. This paper selected a kind of substance from heads of Chinese forest frogs as active fractions, and gave a new path for improving frog product odditional value and realizing synthetically utilizing.Firstly, anticoagulant substances were isolated from the heads of Chinese forest frogs after cutting homogenating, alkali solubilizing, deprotein and drying in this research.. The best extracted technique was examinated by orthogonal tests. The 2 times normal saline was added into Chinese forest frog heads, the alkali treated time was eight hours, the volume of lye was 20 times of the original liquid, the temperature of the alkali solubilizing was 30℃. We did the moderate scale experiment under the best extracted technique and we acquired that the best extracted technique adapted to industrialization.Secondly, the anticoagutant activities of the extracts from the heads of Chinese forest frogs were studied. The extracts could prolong the clotting times, the recalcification times, the prothrombase times, the thrombase times, and inhibit thrombase activities in vitro.The anticoagutant activities were related to the dosages of the extracts from the heads of Chinese forest frogs.The extracts from the heads of Chinese forest frogs also could prolong the bleeding time and the clotting time in vivo,and no relationship with the dosage of the extracts.The anticoagulant efficiency was 5.2U.Then, we also analyzed the physical and chemistry characters of the extracts from the heads of Chinese forest frogs.The extracts were easy to dissove in water, diluent acidic solutions and diluent alkaline solutions except ethanol, aether, ethyl acetate and acetone. They had well heat stability. The anticoagutant activities of the extracts decreased in acid and alkali solutions.The extracts from the heads of Chinese forest frogs were comprised of polysaccharides, proteins, amino acids, hexosamines, galacturonic acids and so on..The active fractions were polysaccharides and proteins. So we concluded that the extracts from the heads of Chinese forest frogs were glycosaminoglycan and heparin-like oligosaccharides.Finally, the anticoagutant mechanisms of the extracts from the heads of Chinese forest frogs were studied. In blood coagulation reaction, the extracts played the anticoagutant role on inhibiting the activities of the blood coagulation factors Ⅱ, Ⅶ, Ⅹ. The active mechanism was similar to heparin. We concluded that the extracts from the heads of Chinese forest frogs were heparin-like oligosaccharides.The conclusions of this research were that the extracts from the heads of Chinese forest frogs were kind of proteoglycan which displayed on the anticoagutant activities in vitro and in vivo. The extracted technique in lab adapted to industrialization. The anticoagulant efficiency was 5.2U. The extracts could inhibit the activities of the blood coagulation factors Ⅱ ,Ⅶ, Ⅹ.
