Objective To investigate the changes and significance of plasma and myocardial ad-renomedullin (ADM) ,endothelin(ET) ,atrial natriuretic peptide(ANP) in pressure overload rats with hypertrophy and the effect and possible mechanism of taurine. Methods 30 Wist-ar rats were randomly divided into there groups, sham - operated group , partial abdominal arota constriction group and partial abdominal arota constriction + taurine - treated group. Systolic blood pressure (SBP) and body weight (BW) was measueed , and left ventricular weight (LVW) was weighted by electronic baiance, left ventricular mess index (LVW/BW) were calculated after six weeks and plasma and myocardial ADM, ET, ANP levels were determined by radioimmunoassay. The myocardial tissue structure was observed under the microscope. Result As compared with sham - operated group , SBP and LVW/BW of partial abdominal arota constriction group were increased by 61. 7% and 19.3%(P<0.05), and plasma ADM,ET ,ratio of ET/ADM and ANPwere increased by 58. 5% , 11. 0% ,31. 5% and 71. l%(P<0.05) respectively,and myocardial ADM,ET ,ratio of ET/ADM and ANPwere increased by 72. 0% , 103. 0% ,43.4% , 18. 3% (P<0. 05). Contrast to partial abdominal arota constriction group , SBP and LVW/BW of partial abdominal arota constriction + taurine - treated group were decreased by 20. 7% , 10. 9 % , plasma ADM , ET , ratio of ET/ADM and ANP were decreased by19.0% , 35. 2% , 21. 3% , 28. 9% (P< 0. 05) , and myocardial ADM,ET .ratio of ET/ADM and ANP decreased by23.1% ,42.0% ,23.3% ,9. 0%(P<0.05)。 There was a positive correlation between plasma and myocardial ADM,ET, ANP and left ventricular mess index , so did ADM and ET, ANP. Conclusion (1) ADM was probably a circulating hormone . In addition, It acted as a local autocrine and/or parac-rine hormone.
