Objective:To study the effects of Chinese herbs Jiu-Ke-Le (JKL) on transforming grows factorβ1 (TGFβ1)anda-smooth muscle actin(α-SMA) in pancreatic tissues in experimental animals and the relationship between alcohol-drinking and pancreatic, we establish the rat model of chronic pancreatitis (CP). This research tried to study and analyse the mechanism of preventing and treating CP. Methods:Male Wistar rats weighing 180-210g were randomly divided into 6 groups: Group A : JKL intragastric administration treatment, Group B: pathology model group, Group C: Daji treatment group, Group D: normal control group, to study the relationship between alcohol-drinking and pancreatic, group E and group F were given intraperitoneal injection of DDC and were perfused stomach with alcohol, Group E: JKL intragastric administration treatment, Group F: without JKL intragastric administration treatment. Each group had 16 rats. Group E and Group F were perfused stomach with alcohol of 8g/kg every day. Except group D, the other rats were used as pathology models of CP. They were induced by intraperitoneal injection of 750mg/kg body weight(BW) of diethyldithiocarbama te (DDC) twice a week for 4 weeks. During the experiment periods,the appetite, water volume, urine volume, stool character should be observed, and weigh the rats once a week. After 4 weeks, the control group and the pathology model one were perfused stomach with saline, 10ml/kg BW. JKL intragastric administration treatment group 10ml/kg BW(2g/kg) and Daji treatment group 10ml/kg BW(50mg/kg),once a day, lasting 4 weeks. The day before the last day, fasting 24 hours, sampled blood about 2 ml in rat's eyeball .Immediately the rats were put to death. Then got out the rats pancreas for the research of immunohistochemistry to measure the expression of TGFβ1 anda-SMA and for the observation by tramission microscope. Results:The CP rats' common conditions of group A C E were obviously better than that of model group B F. The body weight of group A E were better than than that of model group B F (P<0.05) . The levels of blood glucose of A E were lower than that of model group B F (P<0.05) . The levels of amylase of A E were higher than that of model group B, F (P<0.05) JKL was able to decrease the expression of transforming grows factorβ1 and α-smooth muscle actin in pancreatic tissues (P<0.05) . Conclusion:The models of chronic pancreatitis rats can be induced within 4 weeks by intraperitoneal injection of DDC twice a week.JKL can not only improve the common conditions of chronic pancreatitis rats ,decrease the levels of blood glucose ,improve the levels of amylase ,but also can decrease the expression of... |
