Huperzine A Bioadhesive Sustained-release Tablets

Gastrointestinal bioadhesion drug delivery system ( GBDDS ) can be considered as an application of bioadhesion technology to the oral drug delivery system. The paper discussed the thinking and the pathway of the manufacture of sustained-release tablet in the form of GBDDS based on Huperzine A( HupA) , the intellectual property rights of which is possesed by our institute , under the guidance of biopharmaceutical , physical pharmaceutical and pharmacokinetics.HupA , extracted and separated from Chinese herb by Chinese researchers , is a high performance, reversible, high selective Cholinesterase Inhibitor . The thesis adopted HPMC and Carbopol as the main bioadhesion material , aimed to develop once-a-day sustained tablets. We carried out the preformulation study in incision of sustained release and bioadhension and followed the introduction of pharmaceutical necessity. On the basis of the establishment of analytical method, the basic prescription and preparation technology was initially defined by the study of mono-factor experiment and then by the study of formulation screening in the use of orthogonal project and technology optimizing, based on the pharmacy rule as indicator and the stability maintaining of Huperzine A sustained release tablets. The initial stability study show light temperature and humidity have not obvious effects on the HA sustained release tablets, the content and release of the tablets also have not marked change . The result gave a clue that the HA sustained tablet should be packed in moisture proof material and preserved under dry enviroment. The pharmacokinetics and bioavailability study of the HA sustained tablets in beagle dog was made, and the results showed the C_max decreased remarkably, the cure of concentration-time was flat relatively , and character of sustained release was prominent compared with the ordinary tablets.
