.1. Indole Alkaloids Spirtryprostatin B Total Synthesis Of Bisindole Compounds Of Methane Preparation Method Studies

This dissertation includes mainly the following two sections: the total synthetic study of spirotryprostatin B and synthesis of bisindolylkanes.1) Spirotryprostatin B, a potential anti-cancer indole alkaloid, was isolated in 1996 by Osada et al., from the fermentation broth of Aspergillus fumigatus. This section mainly focused on the stereoselective synthetic studies of spirotryprostatin B and its analogues.The strategy toward the synthesis of spirotryprostatin B is focused on MgI2-catalyzed intramolecular annulation reaction of cyclopropanyl spiroox--indole and imine. Starting with the readily available isatin, two designated key cyclization precurors 25 and 26 were synthesized respectively by 5 steps and 6 steps reactions ,with Wittig reaction and Corey-Chaykorsky reaction as key steps. Attempts for the designed intramolecular annulation reaction of cyclopropanyl spirooxindole were reported. At the same time, the effects of the side of cyclopropanyl spirooxindole in MgI2-catalyzed intramolecular annulation reaction were studied as model reactions.All of these results are potentially helpful for the synthesis of the target molecular.2) Bisindolylalkane is an important class of bioactive metabolite. A seriesof bisindolylalkanes were synthesized by nucleophilic addition reactions of indole with N-tert-butanesulfinyl aldimines in moderate to high yields (44%-96% ) catalyzed either by iodine, potassium hydrogen sulfate and amberlyst.Amberlyst is found to catalyze efficiently the electrophilic substitution reactions of indole or substituted indoles with aldehydes in CH3CN under room temperature to afford the corresponding bisindolyalkanes in good to high yields (61%-99%).The structures of all new compounds and key intermediates were established spectroscopically.
