Object: To investigate the influence of traditional Chinese medicine (TCM) for bushenhuoxue (补肾活血) on the pathologic damages of visual pathway in experimental diabetic rats and discuss its effect mechanisms.Methods: The model of experimental diabetic rats was induced by streptozotocin (STZ) .The rats were divided into 7 groups randomly : normal control group, negative control group (model group) , positive control group I , positive control group II and three TCM groups, including low dose, middle dose and hige dose. Expression of glial fibrillary acidic protein (GFAP), Myelin basic protein(MBP), neurotrophic-3(NT-3)in retina, exteral geniculate body (EGB) and visual cortex (VC) was shown by immunohistochemical staining, and was quantitated by Mias-2000 image analysis system.Results: (1)At the 6 months of diabetic course, the positive staining of GFAP, MBP, NT-3 in retina, EGB and VC was reduced to a greater or lesser degree compared with the normol control group (P<0.05 or P<0.01) . (2)Compared with the negative control group, the positive staining of GFAP, MBP, NT-3 in retina , EGB and VC was upregulated in all of the TCM groups, especially in the hige dose group (P<0.05 or P<0.01). (3)By using the same dose of jiangtangling (降糖灵 ) ,the positive stainning of GFAP, MBP, NT-3 in the high dose TCM group was obviously increased compared with the positive control group I (P<0.05 or P<0.01) .Conclusion: after continuous hyperglycemia for 6 months, the experimental diabetic rats showed immunohistochemical changes of neurons and glial cells in visual pathway. The TCM for bushenhuoxue could induce astroglias to generate reactive gliosis in visual pathway, promote recovery of myelin structures, and upregulate the expression of neurotrophic factors. Accordingly, it reduce the pathological damages of neurons and improve the conduct function of visual pathway. It may be one of the important mechanisms of the TCM for bushenhuoxue to relieve the damages of optical function caused by diabetes.
