Objective: Explore the potential superiority of amnion implant in glaucomafiltering surgery by constructing animal glaucoma filtering surgery model .Tosupply the experimental basis for clinic use of amniotic membrane in glaucomafiltering surgery.Method:30 white rabbits is divided evenly into 3 groups atrandom. A group is blank group without operating,B group is operated withglaucoma filtering surgery ,C group is operated with glaucoma filtering surgeryand amnion implant.After 2 weeks of surgery ,we observe the cell cycle offibroblast cell ,stain the filtering pathway tissue with Masson-Trichrome, andexamine it by light microscopy,also observe the filtering pathway tissue byelectronmicroscope. Results: Flow cytometry (FCM) indicate that fibroblast cellof B group occupying proliferation stage of G2/M,however,C group occupyinginhibitory state of G1/S,and cell proliferation index in all groups: B>A>C,P<0.01.light microscopy and electronmicroscope all indicate that fibroblast cellsand collagen fibers in B group are obviously more than which in A group.finestructure of fibroblast cell manifest that B Group is occupying proliferationstate,C group is occupying inhibitory state,A group is occupying normalstate.Conclusion: In conjuctiva healing reaction represented by proliferation offibroblast cell,glaucoma filtering surgery with amnion implant compared withtraditional glaucoma filtering surgery is obviously inhibited.Hence,use ofamnion implant in glaucoma filtering surgery possess major potential.
