Purpose: To observe the effects of the hydroxycamptothecin on cell proliferation and apoptosis of the human lung cancer cell line A549 , to clarify the mechanism, to explore the best concentration of hydroxycamptothecin and to offer reasonable experiment evidence in the clinic therapy.Method: MTT is used to measure the suppress rate of tumor cell, and the apoptosis is observed by electron microscope and light microscope, and the immunohisto chemistry method is used to detect the amount of TGF—β1 and Smad4 of the human lung cancer cell lineA549.Result: (1) the hydroxycamptothecin can suppress the cell proliferation of the human lung cancer cell line A549. The effect was remarkable at the concentration of 100 μ g/ml after 48h, and the inhibitory rate was 63.65%. (2) The lung cancer cells had been observed an inducement on apoptosis by electron microscope and light microscope. (3) The immunohisto chemistry method showed that the hydroxycamptothecin has reduced the amount of TGF—β1 and increased the amount of Smad4 of the human lung cancer cell line A549.Conclusion: (1) The hydroxycamptothecin has the function of inhibiting the proliferation of the human lung cancer cell line A549. (2) The inhibitory effect of hydroxycamptothecin was the most significant when the concentration of hydroxycamptothecin was up to 100μg/ml. (3) The hydroxycamptothecin can induce the apoptosis of the human lung cancer cell line A549. (4) The hydroxycamptothecin can reduce the amount of TGF—β1 and increase the amount of Smad4 of the human lung cancer cell line A549. (5) The hydroxycamptothecin's important mechanism of resisting cancer is inducing the apoptosis and affecting the amount of TGF—β1 and Smad4 of the human lung cancer cell line A549.
